      Uji In Vitro Ekstrak Daun Alpukat (Persea americana Mil.) Terhadap Urolith Kura-kura Sulcata (Centrochelys sulcata)

      Abstract
      Urolithiasis merupakan salah satu gangguan saluran kemih yang sering terjadi pada kura-kura sulcata (Centrochelys sulcata). Penelitian ini bertujuan mengidentifikasi komposisi urolith serta mengevaluasi pengaruh ekstrak daun alpukat (Persea americana Mill.) terhadap peluruhan urolith secara in vitro. Komposisi urolith dianalisis menggunakan Fourier Transform Infrared Spectroscopy (FTIR), sedangkan peluruhan kalsium (Ca) dan magnesium (Mg) dianalisis menggunakan Atomic Absorption Spectrophotometry (AAS). Hasil FTIR menunjukkan bahwa urolith merupakan urolith campuran yang tersusun atas urat, kalsium oksalat, dan fosfat. Pengujian dilakukan dengan menggunakan tiga dosis yaitu 100, 200 dan 300 ppm. Pengukuran kadar kalsium pada semua kelompok perlakuan menunjukkan hasil yang tidak berbeda nyata (p>0,05). Pengukuran kadar magnesium pada dosis 200 ppm menunjukkan hasil yang berbeda nyata (p,0,001). Berdasarkan hasil penelitian, ekstrak daun alpukat belum menunjukkan efektivitas terhadap peluruhan kalsium, tetapi berpotensi meningkatkan peluruhan magnesium pada urolith kura-kura sulcata secara in vitro.
       
      Urolithiasis is one of the most common urinary tract disorders in sulcata tortoises (Centrochelys sulcata). This study aimed to identify the composition of uroliths and evaluate the effect of avocado (Persea americana Mill.) leaf extract on in vitro urolith dissolution. Urolith composition was analyzed using Fourier Transform Infrared Spectroscopy (FTIR), while calcium (Ca) and magnesium (Mg) dissolution was determined using Atomic Absorption Spectrophotometry (AAS). FTIR analysis revealed that the uroliths were mixed uroliths composed of urate, calcium oxalate, and phosphate. The experiment was conducted using three doses of avocado leaf extract: 100, 200, and 300 ppm. Measurement of calcium concentrations in all treatment groups showed no statistically significant differences (p>0.05). Meanwhile, magnesium concentration at the 200 ppm concentration showed a statistically significant difference (p<0,001). Based on the results, avocado leaf extract did not demonstrate effectiveness in calcium dissolution but showed potential to enhance magnesium dissolution in sulcata tortoise uroliths in vitro.
