      Pengaruh Ekstrak Daun Taxus sumatrana terhadap Biomarker Metabolisme Energi pada Mencit ddY Jantan Hiperglikemik

      Abstract
      Hiperglikemia merupakan kondisi yang berdampak luas pada kesehatan manusia dan hewan, dengan prevalensi global mencapai 634,8 juta jiwa pada tahun 2024. Pengobatan berbasis herbal terus dikembangkan sebagai alternatif, salah satunya melalui pemanfaatan daun Taxus sumatrana yang mengandung senyawa flavonoid aktif. Penelitian ini bertujuan mengevaluasi pengaruh ekstrak daun T. sumatrana terhadap biomarker metabolisme energi mencit ddY jantan hiperglikemik. Mencit dibagi dalam 5 kelompok: K1 (5 mg/kg BB), K2 (300 mg/kg BB), K3 (1000 mg/kg BB), K4 (kontrol hiperglikemik), dan K5 (kontrol normal). Hiperglikemia diinduksi menggunakan streptozotocin (STZ) 55 mg/kg BB secara intraperitoneal. Ekstrak diberikan secara oral setiap hari selama 5 minggu. Kadar insulin dan AMPK berbeda nyata antarkelompok (p<0,05). Kelompok K1 menunjukkan kadar insulin 167,33±9,25 mIU/mL, tidak berbeda nyata dengan K3 dan K4, namun nyata lebih tinggi dibandingkan K2 (117,80±13,08 mIU/mL) maupun K5 (80,50±15,93 mIU/mL). Kadar AMPK tertinggi ditunjukkan oleh K2 (28,77±2,97 ng/mL) dan K4 (27,14±0,47 ng/mL), sedangkan K3 (4,48±1,47 ng/mL) dan K5 (8,08±2,89 ng/mL) menunjukkan kadar AMPK terendah, sehingga aktivasi AMPK tidak mengikuti pola dosis-respons linear. Kelompok K1 tetap menunjukkan dinamika berat badan yang relatif stabil (37,72±5,43 gram) serta kadar glukosa darah yang cenderung tidak meningkat (301,27±196,21 mg/dL) dibandingkan kelompok hiperglikemik lainnya.
       
      Hyperglycemia is a condition with widespread impacts on human and animal health, reaching a global prevalence of 634.8 million in 2024. Herbal treatments continue to be developed as alternatives, including the use of Taxus sumatrana leaves containing active flavonoid compounds. This study evaluated the effect of T. sumatrana leaf extract on energy metabolism biomarkers in hyperglycemic male ddY mice. Mice were divided into five groups: K1 (5 mg/kg BW), K2 (300 mg/kg BW), K3 (1000 mg/kg BW), K4 (hyperglycemic control), and K5 (normal control). Hyperglycemia was induced using streptozotocin 55 mg/kg BW intraperitoneally. Extract was administered orally daily for 5 weeks. Insulin and AMPK levels differed significantly between groups (p<0.05). Group K1 showed insulin levels of 167.33±9.25 mIU/mL, not significantly different from K3 and K4, but significantly higher than K2 (117.80±13.08 mIU/mL) and K5 (80.50±15.93 mIU/mL). The highest AMPK levels were shown by K2 (28.77±2.97 ng/mL) and K4 (27.14±0.47 ng/mL), while K3 (4.48±1.47 ng/mL) and K5 (8.08±2.89 ng/mL) were lowest, indicating non-linear dose-response AMPK activation. Group K1 maintained relatively stable body weight of 37.72±5.43 g and non-increasing blood glucose levels of 301.27±196.21 mg/dL compared to other hyperglycemic groups.
