      Perilaku Makan Pentalonia nigronervosa Viruliferus dan Non-viruliferus pada Tanaman Pisang Sehat dan Terinfeksi BBTV Berdasarkan Metode EPG

      Abstract
      Banana bunchy top virus (BBTV) merupakan salah satu patogen virus paling merugikan pada tanaman pisang dan ditularkan terutama oleh kutudaun pisang, Pentalonia nigronervosa. Meskipun perilaku makan vektor ini telah banyak dipelajari, bagaimana status infeksi tanaman dan status viruliferus kutudaun berinteraksi dalam membentuk perilaku makan vektor masih belum dipahami dengan jelas. Penelitian ini mengkaji perilaku makan P. nigronervosa viruliferus dan non-viruliferus pada tanaman pisang Cavendish sehat dan terinfeksi BBTV menggunakan teknik electrical penetration graph (EPG). Penelitian menggunakan rancangan faktorial 2 × 2 dengan perekaman perilaku makan selama 10 jam. Aktivitas penetrasi awal stilet tidak dipengaruhi secara nyata oleh status infeksi tanaman maupun status viruliferus kutudaun. Sebaliknya, interaksi kedua faktor tersebut memengaruhi aktivitas makan pada jaringan vaskular. Kutudaun non-viruliferus pada tanaman terinfeksi BBTV memiliki durasi ingesti floem paling panjang, yang berpotensi meningkatkan peluang akuisisi virus. Sebaliknya, kutudaun viruliferus pada tanaman terinfeksi BBTV menunjukkan durasi non-probing dan ingesti xilem yang lebih panjang, disertai penurunan aktivitas salivasi dan ingesti floem. Hasil ini menunjukkan bahwa status infeksi tanaman dan status viruliferus kutudaun terutama memodifikasi perilaku makan setelah stilet mencapai jaringan vaskular, dengan respons makan yang berbeda sebelum dan setelah virus diperoleh. Penelitian ini memberikan dasar biologis untuk memahami perilaku makan P. nigronervosa selama proses akuisisi BBTV dan setelah kutudaun memperoleh virus.
       
      Banana bunchy top virus (BBTV) is one of the most destructive viral pathogens of banana and is transmitted primarily by the banana aphid, Pentalonia nigronervosa. Although the feeding behavior of this vector has been widely studied, how plant infection status and aphid viruliferous status interact to shape vector feeding behavior remains poorly understood. This study investigated the feeding behavior of viruliferous and non-viruliferous P. nigronervosa on healthy and BBTV-infected Cavendish banana plants using the electrical penetration graph (EPG) technique. The experiment followed a 2 × 2 factorial design, with feeding behavior recorded for 10 h. Early stylet penetration was not significantly affected by plant infection status or aphid viruliferous status. In contrast, the interaction between the two factors affected vascular feeding activities. Non-viruliferous aphids on BBTV-infected plants exhibited the longest duration of phloem ingestion, potentially increasing the opportunity for virus acquisition. Conversely, viruliferous aphids on BBTV-infected plants showed longer durations of non-probing and xylem ingestion, accompanied by reduced phloem salivation and phloem ingestion. These results indicate that plant infection status and aphid viruliferous status primarily modify feeding behavior after stylet penetration into vascular tissues, with distinct feeding responses before and after virus acquisition. This study provides a biological basis for understanding the feeding behavior of P. nigronervosa during BBTV acquisition and after the aphid acquires the virus.
