      Respons Olfaktori Microplitis manilae (Hymenoptera: Braconidae) terhadap Senyawa Volatil Tanaman akibat Berbagai Jenis Pelukaan

      Abstract
      Spodoptera litura dan Spodoptera frugiperda merupakan hama utama tanaman jagung (Zea mays) yang mampu menginduksi pelepasan senyawa volatil sebagai isyarat kimia dalam interaksi tritrofik dengan parasitoid larva Microplitis manilae. Penelitian ini bertujuan mengevaluasi respons olfaktori M. manilae terhadap volatil tanaman jagung akibat berbagai jenis pelukaan serta mengidentifikasi profil senyawa volatil yang dihasilkan. Respons olfaktori diuji menggunakan olfaktometer empat lengan dengan lima sumber volatil, yaitu tanaman sehat, tanaman dengan pelukaan mekanis, tanaman terinfestasi S. litura, tanaman terinfestasi S. frugiperda, dan udara bersih sebagai kontrol, dengan waktu residensi parasitoid sebagai peubah respons yang dianalisis menggunakan linear mixed-effects models (LMMs) dan estimated marginal means. Profil senyawa volatil dikoleksi menggunakan adsorben karbon aktif dan dianalisis dengan gas chromatography-mass spectrometry (GC-MS). Hasil penelitian menunjukkan bahwa M. manilae memberikan respons olfaktori yang nyata terhadap volatil tanaman jagung dibandingkan udara bersih (P < 0,001), serta menunjukkan waktu residensi yang lebih lama pada tanaman terinfestasi S. litura maupun S. frugiperda dibandingkan tanaman sehat dan tanaman dengan pelukaan mekanis, tanpa perbedaan nyata di antara kedua spesies Spodoptera tersebut (P > 0,05). Analisis GC-MS menunjukkan peningkatan jumlah dan intensitas senyawa volatil seiring meningkatnya tingkat infestasi herbivora, dengan profil paling kompleks (tujuh senyawa) ditemukan pada tanaman terinfestasi S. frugiperda. Senyawa yang teridentifikasi berupa taurine, n-methyltaurine, carbonyldiamide, dan sekelompok alkana rantai pendek (isobutane, n-butane, iso-pentane, tert-butyl alcohol) berbeda dari golongan terpenoid yang umum dilaporkan sebagai herbivore-induced plant volatiles (HIPVs), sehingga perannya sebagai kairomon masih perlu dikonfirmasi melalui analisis lanjutan. Secara keseluruhan, hasil penelitian ini menunjukkan bahwa volatil tanaman jagung berperan penting dalam memediasi perilaku pencarian inang M. manilae serta mendukung potensinya sebagai agens pengendali hayati terhadap S. litura dan S. frugiperda pada agroekosistem jagung.
       
      Spodoptera litura and Spodoptera frugiperda are major insect pests of maize (Zea mays) that induce the release of volatile compounds acting as chemical cues in the tritrophic interaction with the larval parasitoid Microplitis manilae. This study aimed to evaluate the olfactory responses of M. manilae to maize volatiles induced by different types of damage and to characterize the resulting volatile profiles. Olfactory responses were tested using a four-arm olfactometer with five odor sources: healthy plants, mechanically damaged plants, S. litura-infested plants, S. frugiperda-infested plants, and clean air as control, with parasitoid residence time as the response variable, analyzed using linear mixed-effects models (LMMs) and estimated marginal means. Volatile profiles were collected using activated-charcoal adsorbent and analyzed by gas chromatography-mass spectrometry (GC-MS). Microplitis manilae showed a significantly stronger olfactory response to maize volatiles than to clean air (P < 0.001), with longer residence times on plants infested by either S. litura or S. frugiperda compared with healthy or mechanically damaged plants, but with no significant difference between the two Spodoptera species (P > 0.05). GC-MS analysis showed that the number and intensity of volatile compounds increased with the level of herbivore infestation, with the most complex profile (seven compounds) found in S. frugiperda-infested plants. The identified compounds—taurine, n-methyltaurine, carbonyldiamide, and a group of short-chain alkanes (isobutane, n-butane, iso-pentane, tert-butyl alcohol)—differ from the terpenoid classes commonly reported as herbivore-induced plant volatiles (HIPVs), and their role as kairomones therefore requires further confirmation. Overall, these findings indicate that maize volatiles play an important role in mediating host-searching behavior in M. manilae and support its potential as a biological control agent against S. litura and S. frugiperda in maize agroecosystems.
