      Ekstraksi dan Karakterisasi Kitosan dari Shedding Lalat Tentara Hitam Hermentia illucens dengan Metode Microwave-Assisted Extraction

      Abstract
      Kitosan merupakan biopolimer turunan kitin yang diaplikasikan secara luas dalam berbagai bidang. Kepompong (shedding) lalat tentara hitam (Hermetia illucens atau BSF) berpotensi sebagai sumber kitin alternatif yang belum dimanfaatkan secara optimal. Penelitian ini bertujuan mengekstraksi kitosan dari shedding BSF menggunakan metode Microwave-Assisted Extraction (MAE) pada tahap deasetilasi dan mengidentifikasi pengaruh daya (200 W dan 400 W) serta waktu (10, 15, dan 20 menit) terhadap karakteristik absorbansi sinar UV-Vis kitosan yang dihasilkan. Hasil penelitian menunjukkan bahwa penggunaan metode MAE berhasil mempersingkat waktu ekstraksi secara signifikan. Berdasarkan analisis absorbansi, sampel B (200 W, 15 menit) dan D (400 W, 10 menit) menghasilkan nilai absorbansi tertinggi, namun masih terindikasi adanya pengotor berupa sisa protein pada puncak 280 nm. Sampel A (200 W, 10 menit) dan E (400 W, 15 menit) dinilai sebagai sampel yang paling ideal karena profil absorbansinya tidak menunjukkan adanya indikasi pengotor meskipun memiliki nilai absorbansi yang lebih rendah. Sebaliknya, proses deasetilasi selama 20 menit (sampel C dan F) menghasilkan absorbansi terendah yang mengindikasikan bahwa perlakuan waktu yang terlalu lama bersifat destruktif terhadap struktur kitosan.
       
      Chitosan is a chitin-derived biopolymer widely applied in various fields. Shedding from the black soldier fly (Hermetia illucens, BSF) has the potential to act as an alternative chitin source that remains underutilized. This study aimed to extract chitosan from BSF shedding using the Microwave-Assisted Extraction (MAE) method during the deacetylation stage and to identify the effect of microwave power (200 W and 400 W) and time (10, 15, and 20 minutes) on the UV-Vis absorbance characteristics of the produced chitosan.. The results showed that the MAE method successfully and significantly shortened the extraction time. Based on the absorbance analysis, samples B (200 W, 15 minutes) and D (400 W, 10 minutes) yielded the highest absorbance values but still indicated the presence of impurities in the form of residual protein at the 280 nm peak. Samples A (200 W, 10 minutes) and E (400 W, 15 minutes) were considered the most ideal samples as their absorbance profiles showed no indication of impurities, despite having lower absorbance values. Conversely, a deacetylation time of 20 minutes (samples C and F) resulted in the lowest absorbance, indicating that prolonged treatment time has a destructive effect on the chitosan structure.
