Vanilla planifolia merupakan tanaman rempah paling berharga kedua secara global setelah saffron dan bernilai ekonomi karena kandungan senyawa vanilin pada polongnya. Perbanyakan vanili menggunakan stek batang dinilai kurang efisien karena membutuhkan lahan luas, waktu lama, dan bahan tanam lebih banyak. Permasalahan lain yaitu viabilitas dan daya kecambah biji vanili yang rendah karena tidak memiliki endosperma. Kultur jaringan menjadi salah satu alternatif untuk memproduksi bibit vanili secara cepat, seragam, dan bebas patogen. Organogenesis vanili, khusus melalui induksi kalus, juga dapat dimanfaatkan untuk mendapatkan keragaman genetik melalui variasi somaklonal. Kalus hasil induksi dapat diarahkan kepada beberapa tujuan lain seperti induksi mutasi, kultur suspensi sel, kultur protoplas, dan transformasi gen. Penelitian ini bertujuan mempelajari pengaruh jenis eksplan dan komposisi ZPT terhadap induksi dan regenerasi kalus serta multiplikasi tunas vanili varietas Vania I secara in vitro.
Percobaan 1 merupakan percobaan organogenesis vanili tidak langsung
melalui induksi kalus. Percobaan ini terdiri atas 3 sub-percobaan. Percobaan 1a dan
1b merupakan percobaan induksi kalus vanili yang disusun dalam Rancangan
Kelompok Lengkap Teracak (RKLT) dua faktor. Sub-percobaan 1a menggunakan
eksplan ruas, sedangkan sub-percobaan 1b menggunakan eksplan daun yang
diperoleh dari planlet vanili in vitro varietas Vania 1 untuk induksi kalus. Faktor
pertama pada masing-masing sub percobaan adalah posisi eksplan (atas, tengah,
dan bawah). Posisi eksplan atas diambil dari ruas atau daun pada buku ke 1-2 di
bawah pucuk; eksplan tengah diambil dari bagian antara buku ke 3-4; dan eksplan
bawah diambil dari bagian antara buku ke-5 sampai basal planlet. Faktor kedua
adalah komposisi zat pengaruh tumbuh: kontrol tanpa ZPT (Z0); 4,52 µM 2,4-D +
2,27 µM TDZ (Z1); 9,04 µM 2,4-D + 2,27 µM TDZ (Z2); 4,52 µM picloram + 2,27
µM TDZ (Z3); 9,04 µM picloram + 2,27 µM TDZ (Z4); 9,04 µM NAA + 9,04 µM
BAP (Z5); dan 9,04 µM TDZ (Z6). Percobaan induksi dilakukan dengan tiga
tahapan kerja, yaitu persiapan alat dan media tanam, penanaman bahan tanam ke
dalam media induksi kalus, sampai proliferasi kalus.
Proliferasi kalus vanili dilakukan untuk memperbanyak jumlah bahan tanam
dan menyeleksi kalus terbaik untuk diregenerasikan. Eksplan yang digunakan untuk
proliferasi berupa kalus kompak yang berasal dari ruas. Kalus tersebut dipindahkan
ke media proliferasi ½ MS + 4,64 µM kinetin + 0,54 µM NAA dan disubkultur
sebanyak 3 kali dengan interval waktu 1 bulan pada media proliferasi yang sama.
Luas kalus dan perubahan warna kalus dievaluasi pada setiap kali subkultur.
Percobaan 1c merupakan percobaan regenerasi kalus vanili yang diinduksi
dari eksplan ruas. Percobaan ini disusun menggunakan Rancangan Acak Lengkap
(RAL) dengan faktor tunggal yaitu jenis dan konsentrasi sitokinin yang terdiri atas
5 media: kontrol tanpa sitokinin (MS0), 9,30 µM Kinetin, 13,82 µM Kinetin, 8,88
µM BAP, dan 13.32 µM BAP. Kemampuan regenerasi kalus, jumlah tunas, dan
akar vanili pada setiap media regenerasi diamati selama 12 MSK.
Percobaan 2 merupakan multiplikasi tunas aksilar vanili secara in vitro.
Percobaan ini dirancang menggunakan Rancangan Kelompok Lengkap Teracak
(RKLT) dua faktor. Faktor pertama adalah konsentrasi NAA (0; 2,68; dan 8,05
µM), sedangkan faktor kedua adalah konsentrasi BAP (4,44; 8,88; dan 13,32 µM).
Kemampuan multiplikasi eksplan dievaluasi berdasarkan variabel jumlah tunas,
akar, dan daun vanili yang terbentuk selama 12 MSK.
Respons induksi kalus pada percobaan 1a dan 1b menunjukkan bahwa
penggunaan eksplan ruas maupun daun in vitro vanili memberikan pengaruh yang
sama terhadap induksi kalus vanili. Penggunaan eksplan ruas bawah menghasilkan
persentase pembentukan kalus tertinggi (22,9%) dibandingkan ruas atas dan tengah.
Respons induksi kalus vanili tertinggi dalam penelitian ini diperoleh pada
komposisi media MS + 9,04 µM picloram + 2,27 µM TDZ dengan persentase
keberhasilan pembentukan kalus sebesar 22,2% pada eksplan ruas dan 15,0% pada
eksplan daun. Tipe kalus yang diperoleh dalam penelitian ini terbagi menjadi dua
yaitu (i) kalus kompak berwarna putih yang dominan terbentuk dari eksplan ruas
vanili; dan (ii) kalus remah berwarna hijau transparan dari eksplan ruas muda dan
tengah, serta eksplan daun vanili.
Kalus vanili yang berasal dari eksplan ruas dipindahkan ke media proliferasi
½ MS + kinetin + NAA. Ukuran kalus bertambah secara cepat sejak minggu ketiga
pada subkultur I hingga subkultur II. Perubahan warna kalus vanili menuju senesens
mulai terjadi pada subkultur III. Kalus kompak vanili berwarna putih sampai putih
krem dipilih untuk diregenerasikan menjadi tunas. Kalus tersebut dipindahkan ke
media regenerasi yang mengandung sitokinin BAP atau kinetin. Hasil percobaan 1c
menunjukkan bahwa media MS dengan penambahan BAP 8,88 µM menghasilkan
respons regenerasi tunas tertinggi sebesar 50% dan rata-rata jumlah tunas 1,33 tunas
per kalus dengan tinggi 3,18 mm. Penggunaan BAP dinilai lebih efektif
dibandingkan kinetin dilihat dari persentase respons kalus vanili membentuk tunas
maupun akar. Media kontrol MS0 tidak menunjukkan regenerasi tunas, namun
hanya menghasilkan respons pembentukan akar sebesar 8,33% dengan panjang
akar 1,48 mm.
Hasil percobaan 2 menunjukkan bahwa daya multiplikasi tunas vanili
dengan mikrostek dipengaruhi oleh konsentrasi BAP yang digunakan. Faktor
tunggal NAA maupun interaksinya dengan BAP tidak mempengaruhi jumlah tunas
vanili yang terbentuk. Penambahan BAP 4,44 µM ke dalam media MS
menghasilkan rerata jumlah tunas vanili tertinggi yaitu 2,2 tunas per eksplan selama
12 minggu kultur. Jalur multiplikasi tunas vanili menggunakan mikrostek
cenderung lebih efisien karena kemampuan regenerasi tunas lebih cepat
dibandingkan organogenesis tidak langsung melalui induksi kalus. Organogenesis
tidak langsung melalui induksi kalus memiliki keunggulan lain dalam fleksibilitas
penggunaan jenis eksplan serta berpotensi dimanfaatkan dalam berbagai aplikasi
bioteknologi lanjutan pada berbagai spesies tanaman.
Penelitian ini menghasilkan informasi protokol perbanyakan vanili secara in
vitro melalui organogenesis tidak langsung dengan induksi kalus dan multiplikasi
tunas aksilar vanili. Protokol dalam penelitian ini dapat menjadi dasar bagi
penerapan bioteknologi lanjutan seperti mutagenesis, transformasi genetik, atau
produksi senyawa metabolit sekunder untuk menghasilkan kalus atau planlet
sebagai bahan tanam. Informasi protokol multiplikasi tunas dapat digunakan untuk
penyediaan bibit vanili, namun masih membutuhkan informasi tambahan terkait
metode elongasi, perakaran, dan aklimatisasi yang optimal. Penggunaan protokol
induksi dan regenerasi kalus, yang melibatkan ZPT sintetik seperti picloram dan
TDZ pada konsentrasi tinggi, memerlukan pengujian kestabilan genetik sebelum
diterapkan sebagai protokol penyediaan bibit yang seragam. Vanilla planifolia is the second most valuable spice crop worldwide after
saffron, with economic value derived from the vanillin content in its pods.
Conventional propagation through stem cuttings is considered inefficient because
it requires extensive planting area, prolonged production time, and a large amount
of planting material. Another problem is the low viability and germination rate of
vanilla seeds, which lack an endosperm. Plant tissue culture represents an
alternative approach for the rapid production of genetically uniform and pathogen
free vanilla planting materials. In vitro vanilla organogenesis, particularly through
callus induction, can also be utilized to generate genetic diversity through
somaclonal variation. Induced callus can be directed toward other applications such
as mutation induction, cell suspension culture, protoplast culture, and genetic
transformation. This study aims to investigate the effects of explant type and plant
growth regulator (PGR) composition on callus induction and regeneration, as well
as the in vitro multiplication of shoots from the vanilla cultivar vania 1.
Experiment 1 investigated Indirect organogenesis of vanilla through callus
induction and consisted of three sub-experiments. Sub-experiments 1a and 1b
evaluated vanilla callus induction using a two-factor completely randomized block
design (CRBD). Sub-experiment 1a used node explants, while sub-experiment 1b
used leaf explants obtained from in vitro plantlets of vanilla cv. Vania 1. The first
factor in each sub-experiment was the explant position (upper, middle, basal). The
upper explants were collected from internode or leaf at the first and second node
below the shoot apex; middle explants from the third and fourth nodes; and basal
explants from the fifth nodes to the basal region of the mother plantlet. The second
factor was the composition of the growth-promoting agents: control (Z0); 4,52 µM
2,4-D + 2,27 µM TDZ (Z1); 9,04 µM 2,4-D + 2,27 µM TDZ (Z2); 4,52 µM
picloram + 2,27 µM TDZ (Z3); 9,04 µM picloram + 2,27 µM TDZ (Z4); 9,04 µM
NAA + 9,04 µM BAP (Z5); and 9,04 µM TDZ (Z6). The experiment was conducted
in three stages: preparation of the growth medium, planting of the plant material on
the callus induction medium, and callus proliferation.
Vanilla callus proliferation was conducted to increase the amount of
propagative material and to select the most suitable calli for regeneration. Compact
calli derived from internode explant were transferred to a proliferation medium of
½ MS + 4,64 µM kinetin + 0,54 µM NAA and subcultured three times at one-month
intervals. Callus area and color changes were evaluated at each subculture. Sub
experiment 1c evaluated the regeneration of vanilla callus induced from internode
explants. The experiment was arranged in a completely randomized design (CRD)
with a single factor, cytokinin type, consisting of a control medium (MS0), 9,30
µM kinetin, 13,82 µM kinetin, 8,88 µM BAP, and 13,32 µM BAP. Regeneration
responses of callus regeneration, number of shoots, and roots produced were
evaluated over a 12- week culture period.
Experiment 2 investigate the in vitro multiplication of vanilla axillary
shoots. The experiment was arranged in a two factor randomized complete blok
design (RCBD). The first factor was NAA concentration (0; 2.68; and 8.05 µM),
while the second factor was BAP concentration (4.44; 8.88; and 13.32 µM). Explant
multiplication was evaluated based on the number of shoots, roots, and leaves
produced during 12 weeks after culture.
The results of experiments 1a and 1b showed that the use of both node and
leaf explants from in vitro vanilla plantlets resulted in similar callus induction
responses. Basal internode explants produced the highest callus formation
percentage (22.9%) compared to the upper and middle internodes. The highest
callus induction response was obtained on MS medium formulation containing 9.04
µM picloram and 2.27 µM TDZ, with a callus formation percentages of 22.2% for
node explants and 15.0% for leaf explants. Two types of callus were obtained in
this study: (i) compact, white callus, which predominantly formed from vanilla
node explants; and (ii) crumbly, translucent green callus from young and middle
node explants, as well as vanilla leaf explants.
Vanilla calli derived from nodal explant were transferred to proliferation
medium half-strength MS supplement with kinetin and NAA. Callus size increased
rapidly from the third week during subculture I through subculture II. Changes in
callus color associated with senescence began to occur during the subculture III.
Compact white to cream-white calli were selected for shoot regeneration. The
selected calli were transferred to regeneration media containing either BAP or
kinetin. The results of sub-experiment 1c showed that MS medium supplemented
with 8,88 µM produced the highest shoot regeneration response (50%), with
average of 1,33 shoots per callus and a mean shoot height of 3,18 mm. BAP was
considered more effective than kinetin in promoting both shoot and root
regeneration. The control medium (MS0) did not induce shoot regeneration but
produced a root formation response of only 8,33$, with an average root length of
1,48 mm.
The results of Experiment 2 showed that the multiplication rate of vanilla
shoots using microcuttings was influenced by the concentration of BAP used. NAA
concentration alone and its interaction with BAP did not significantly affected the
number of vanilla shoots formed. The MS medium supplemented with 4.44 µM
BAP was the best medium for in vitro multiplication of vanilla shoots, yielding 2.2
shoots per explant over 12 weeks of culture. The microcutting-based shoot
multiplication pathway tended to be more efficient because shoot regeneration
occured more rapidly than through indirect organogenesis via callus induction.
Nevertheless, indirect organogenesis through callus induction offers advantages in
term of flexibility in explant selection and its potential application in various
downstream biotechnological approaches across plant species.
This study provides an information on in vitro vanilla propagation protocols
through indirect organogenesis via callus induction and axillary shoot
multiplication. The protocols developed in this study may serve as a basis for futher
biotechnological applications, including mutagenesis, genetic transformation, and
secondary metabolite production, using callus or plantlets as starting materials. The
shoot multiplication protocol may utulized for vanilla seedling production.
However, futher optimization of shoot elongation, rooting, and acclimatization is
required. The use of callus induction and regeneration protocols involving syntetic
PGRs such as picloram and TDZ at relatively high concentrations also requires
genetic stability assessment before the protocols can be applied for the production
of genetically uniform planting materials.
