dc.description.abstractUlat grayak (Spodoptera litura) merupakan salah satu hama penting pada pertanian hortikultura yang bersifat polifag dengan tanaman inang lebih dari 40 famili tanaman dan mampu menurunkan produktivitas tanaman secara signifikan. Petani umumnya menggunakan insektisida sintetik untuk mengendalikan S. litura karena dianggap lebih efektif dan memberikan hasil pengendalian yang lebih cepat. Penggunaan insektisida sintetik dengan bahan aktif sama secara berulang dapat memicu perkembangan resistensi, resurjensi hama, ledakan hama sekunder, dan pencemaran lingkungan. Hama S. litura memiliki tingkat kerentanan rendah terhadap profenofos, sehingga berpotensi mengalami resistensi terkait perubahan target-site insektisida. Paparan profenofos berulang dapat menyebabkan perubahan pada enzim asetilkolinesterase (AChE) melalui perubahan genetik maupun regulasi aktivitas enzim yang berpotensi meningkatkan aktivitas AChE dan menurunkan keefektifan insektisida dalam tubuh serangga. Penelitian ini bertujuan menganalisis dan menentukan hubungan antara aktivitas AChE dengan nisbah resistensi S. litura terhadap insektisida profenofos. Metode uji yang digunakan mencakup uji pendahuluan, uji lanjutan, dan induksi resistensi dengan teknik celup daun dan analisis aktivitas AChE pada setiap generasi uji. Aktivitas AChE dianalisis menggunakan metode Ellman. Homogenat larva S. litura instar III disentrifugasi dan supernatan digunakan sebagai sumber enzim. Absorbansi diukur pada 412 nm menggunakan spektrofotometer. Data mortalitas dianalisis dengan analisis probit untuk menentukan lethal concentration 50 (LC50), penentuan tingkat resistensi melalui nisbah resistensi (NR). Hasil penelitian menunjukkan peningkatan status resistensi larva S. litura terhadap insektisida profenofos pada setiap generasi, dan aktivitas enzim AChE berkorelasi positif dengan NR. Temuan penelitian ini menunjukkan bahwa perubahan aktivitas AChE berperan dalam perkembangan resistensi S. litura terhadap profenofos dan dapat menjadi indikator biokimia dalam pemantauan resistensi insektisida. Informasi ini dapat mendukung pengelolaan resistensi melalui strategi penggunaan insektisida yang lebih efektif dan berkelanjutan.
dc.description.abstractThe armyworm (Spodoptera litura) is one of the major pests in horticultural crops, exhibiting polyphagous characteristics with host plants spanning more than 40 plant families and being capable of significantly reducing crop productivity. Farmers generally use synthetic insecticides to control S. litura due to be effective and rapid control results. However, repeated use of synthetic insecticides with the same active ingredient can trigger the development of resistance, pest resurgence, secondary pest outbreaks, and environmental pollution. S. litura has a low susceptibility to profenofos, indicating its potential to develop resistance associated with alterations in the insecticide target site. Repeated exposure to profenofos may cause changes in acetylcholinesterase (AChE) activity through genetic modifications or regulation of enzyme activity, potentially increasing AChE activity and reducing insecticide effectiveness within the insect body. This study aimed to analyze the relationship between AChE activity and the resistance ratio of S. litura to profenofos. The experimental methods included preliminary, advanced, and resistance induction tests using the leaf-dip technique, followed by AChE activity analysis in each test generation. AChE activity was analyzed using the Ellman method. Third-instar larval homogenates of S. litura were centrifuged, and the supernatant was used as the enzyme source. Absorbance was measured at 412 nm using a spectrophotometer. Mortality data were analyzed using probit analysis to determine the lethal concentration 50 (LC50), and resistance levels were determined based on the resistance ratio (RR). The results showed an increase in the resistance status of S. litura larvae to profenofos across generations, and AChE activity was positively correlated with RR. The findings of this study indicate that changes in AChE activity are associated with the development of profenofos resistance in S. litura and can serve as a biochemical indicator for monitoring insecticide resistance. This information can support resistance management through more effective and sustainable insecticide use strategies.
