      Potensi Tumbuhan Hiperakumulator Nikel (Ni) sebagai Agen Fitoremediasi dan Fitomining di Pomalaa, Sulawesi Tenggara

      Abstract
      Indonesia merupakan salah satu hotspot lahan ultramafik terbesar di dunia, sehingga memiliki potensi besar untuk menemukan banyak tumbuhan hiperakumulator, yaitu tumbuhan pengakumulasi logam dalam jumlah besar khususnya nikel. Meski demikian, potensi tersebut belum secara optimal tergali. Oleh karena itu, saat ini data jenis tumbuhan hiperakumulator di Indonesia masih relatif sangat sedikit dibandingkan dengan negara lain yang juga memiliki area ultramafik. Tumbuhan hiperakumulator berpotensi sebagai agen fitoremediasi yaitu upaya pembersihan tingkat kontaminan di lingkungan yang tercemar logam, sekaligus berpotensi sebagai agen fitomining yaitu proses pengambilan logam berharga dari tanah melalui perantara tumbuhan penyerap logam. Eksplorasi dilakukan di kawasan izin usaha pertambangan PT ANTAM Tbk UBP Nikel Kolaka, Kecamatan Pomalaa, Sulawesi Tenggara. Area eksplorasi meliputi area dengan vegetasi lokal berupa hutan primer, fragmentasi dan sekunder. Penelusuran tumbuhan yang berpotensi mengakumulasi nikel diseleksi menggunakan nickel test paper yaitu alat uji semi kuantitatif skala lapangan menggunakan kertas yang telah diresapi dengan dimethylglyoxime (DMG). Ketika kertas tersebut menunjukkan uji positif, maka kertas uji berubah menjadi warna merah atau magenta. Tumbuhan yang menunjukkan reaksi positif pada kertas uji kemudian dikoleksi untuk dilakukan analisis kadar logamnya. Analisis logam dilakukan pada sampel daun dan akar pada tumbuhan serta sampel tanah di sekitarnya. Pengambilan data vegetasi dilakukan di tiga stasiun (12 plot) untuk menilai kelompok tumbuhan adaptif berdasarkan analisis struktur dan komposisi spesies. Analisis data ekologi menggunakan Indeks Nilai Penting (INP), sedangkan data kimia tumbuhan dan tanah dianalisis untuk mengevaluasi kemampuan spesies dalam mengakumulasi dan mentranslokasikan logam berdasarkan perhitungan bioconcentration factor (BCF) dan translocation factor (TF). Hasil analisis kimia tanah menunjukkan bahwa rata-rata kandungan logam Ni, Cr dan Co di tanah melebihi batas kritis logam di tanah industri. Keberadaan logam tersebut ketika terakumulasi dalam jaringan tubuh makhluk hidup akan menyebabkan gejala toksisitas karena tingginya logam di tanah.Tumbuhan yang mampu beradaptasi pada kondisi lingkungan yang ekstrem ini hanyalah kelompok tumbuhan adaptif dengan mekanisme adaptasi yang tinggi terhadap tingkat toleransi logam. Sebanyak 37 famili yang terdiri atas 59 spesies dan 3385 individu, termasuk di dalamnya 5 spesies Endemik Sulawesi berhasil diidentifikasi yang berasal dari tanah ultramafik Pomalaa. Berdasarkan komposisinya, jumlah individu tertinggi pada setiap habitusnya diwakili oleh Xanthostemon contertiflorus pada habitus pohon, Leucosyke capitellata pada habitus semak, Spathoglottis plicata pada habitus herba, Dinochloa sp. pada habitus liana, dan Baumea rubiginosa pada habitus rumput. Berdasarkan struktur komunitasnya, Syzygium sp., Gymnostoma sumatranum, Buchanania arborescens, dan Baumea rubiginosa memiliki INP paling tinggi pada setiap strata pertumbuhan, yakni pohon, tiang, pancang dan semai. Hasil eksplorasi melalui uji DMG dan analisis kimia menunjukkan bahwa dalam penelitian ini ditemukan dua spesies tumbuhan hiperakumulator nikel dengan kandungan nikel pada berat kering daun > 1000 mg/kg yang digunakan sebagai kriteria dasar hiperakumulator Ni yaitu Phyllanthus littoralis dan Macaranga celebica. Kedua spesies ini menunjukkan akumulasi nikel yang tinggi pada bagian daun maupun akarnya, dengan kadar nikel di daun yaitu 4125,41 mg/kg dan 1015,11 mg/kg. Begitu pula dengan kandungan logam pada akar juga menunjukkan akumulasi nikel yang tinggi yaitu sekitar 2782,58 mg/kg dan 1166,56 mg/kg. Temuan ini merupakan catatan terbaru penemuan hiperakumulator nikel di hutan ultramafik Pomalaa, Kabupaten Kolaka, Sulawesi Tenggara. Selain dua spesies yang masuk kategorisasi hiperakumulator nikel pada batas nikel di daun, dalam penelitian ini juga terdapat delapan spesies lain yang memiliki kemampuan akumulasi nikel tinggi di akar (> 1000 mg/kg), meskipun konsentrasi nikel di daun rendah. Kedelapan spesies tersebut adalah Ficus subtinervia, Planchonella obovata, Gironniera subaequalis, Rigiolepsis argentii, Guettardella hexasperma, Leucosyke capitellata, Diospyros ferrea, dan Commersonia bartramia. Selain itu, R. argentii menjadi salah satu spesies endemik Sulawesi dan menjadi catatan terbaru penemuannya di Pomalaa, Sulawesi Tenggara. Oleh karena itu, dalam penelitin ini mengajukan bahwa akumulasi nikel tinggi di akar (> 1000 mg/kg) dapat dikategorikan sebagai tumbuhan hiperakumualator nikel meskipun konsentrasi nikel di daun rendah. Berdasarkan pertimbangan terhadap delapan spesies tersebut, maka pada penelitian ini ditemukan 10 spesies tumbuhan hiperakumulator nikel di wilayah Pomalaa yang sangat potensial untuk dikembangkan sebagai agen fitoremediasi dan fitomining nikel.
       
      Indonesia is one of the world's largest ultramafic hotspots, giving it immense potential for the discovery of numerous hyperaccumulator plants—plants that accumulate large amounts of metals, particularly nickel. However, this potential has not yet been fully explored; as a result, data on hyperaccumulator species in Indonesia remains relatively scarce compared to other countries that also possess ultramafic areas. Hyperaccumulator plants have potential as phytoremediation agents, efforts to clean up contaminant levels in metal-polluted environments—as well as phytomining agents, which involve the extraction of valuable metals from the soil through metal-accumulating plants. Exploration was conducted in the mining business permit area of PT ANTAM Tbk UBP Nikel Kolaka, Pomalaa District, Southeast Sulawesi. The exploration area encompasses regions with local vegetation, including primary, fragmented, and secondary forests. Screening for plants indicated to potentially accumulate nickel was carried out using nickel test paper, a semi-quantitative field testing tool using paper impregnated with dimethylglyoxime (DMG). A positive test result causes the paper to turn red or magenta. Plants exhibiting a positive reaction on the test paper were then collected for metal content analysis. Metal analysis was performed on leaf and root samples, as well as surrounding soil samples. Vegetation data were collected across tree stations (12 plots) to assess adaptive plant groups based on structural and species composition analysis. Ecological data were analyzed using the Important Value Index (IVI), while plant and soil chemical data were analyzed to evaluate the species' ability to accumulate and translocate metals based on the calculation of the bioconcentration factor (BCF) and translocation factor (TF). Soil chemical analysis revealed that the average concentration of heavy metals (Ni, Cr, and Co) in the soil exceeded critical threshold limits for industrial soils. Accumulation of these metals within the tissues of living organisms causes toxicity symptoms due to the elevated metal concentrations in the soil. Only adaptive plant groups with high metal-tolerance mechanisms are capable of adapting to these extreme environmental conditions. A total of 37 families comprising 59 species and 3,385 individuals, including 5 species endemic to Sulawesi, were identified from the Pomalaa ultramafic soil. Based on species composition, the highest number of individuals across their respective growth habits represented by Xanthostemon confertiflorus among trees, Leucosyke capitellata among shrubs, Spathoglottis plicata among herbs, Dinochloa sp. among lianas, and Baumea rubiginosa among grasses. Furthermore, based on community structure, Syzygium sp., Gymnostoma sumatranum, Buchanania arborescens, and Baumea rubiginosa exhibited the highest Important Value Index (IVI) values across all growth strata: trees, poles, saplings, and seedings. Exploration results utilizing the DMG test and chemical analysis revealed the discovery of two nickel hyperaccumulator species meeting the fundamental criterion for Ni hyperaccumulators—a dry-weight leaf nickel concentration exceeding 1,000 mg/kg: Phyllanthus littoralis and Macaranga celebica. Both species exhibited high nickel accumulation in both leaves and roots, with leaf nickel concentrations of 4,125.41 mg kg-1 and 1,015.11 mg kg-1, respectively. Similarly, root metal content demonstrated high nickel accumulation, reaching approximately 2,782.58 mg kg-1 and 1,166.56 mg kg-1. These findings represent a new record of nickel hyperaccumulator discovery in the ultramafic forest of Pomalaa, Kolaka Regency, Southeast Sulawesi. In addition to the two species categorized as nickel hyperaccumulators based on leaf nickel threshold criteria, in this study also identified eight other species possessing high nickel accumulation capacity in their roots (> 1,000 mg kg-1) Ficus subtinervia, Planchonella obovata, Gironniera subaequalis, Rigiolepis argentii, Guettardella hexasperma, Leucosyke capitellata, Diospyros ferrea, and Commersonia bartramia. Furthermore, R. argentii represents an endemic species to Sulawesi, marking a new record of its occurrence in Pomalaa, Southeast Sulawesi. Therefore, in this study propose that high nickel accumulation in roots (> 1,000 mg kg-1) can be categorized as nickel hyperaccumulation, even when leaf nickel concentration is low. Considering these eight species, this study identified a total of ten nickel hyperaccumulator plant species in the Pomalaa region, which hold significant potential for development as agents in phytoremediation and nickel phytomining.
