      Microneedle Berbahan Hidroksiapatit untuk Sistem Penghantaran Obat Klindamisin

      Abstract
      Infeksi kulit oleh bakteri Staphylococcus aureus yang resisten terhadap antibiotik menjadi permasalahan yang memerlukan sistem penghantaran obat yang lebih efektif dibandingkan metode konvensional seperti oral dan topikal. Penelitian ini bertujuan mengetahui karakteristik fisika microneedle (MNs) berbahan hidroksiapatit (HAp) serta pelepasan klindamisin dalam sistem penghantarannya berdasarkan variasi formulasi. HAp disintesis dari cangkang telur ayam melalui kalsinasi dan reaksi dengan asam fosfat, kemudian dikarakterisasi menggunakan XRD. Komposit Gel/HAp yang dicampur klindamisin dan glutaraldehida dicetak menjadi MNs menggunakan cetakan PDMS, lalu dikarakterisasi morfologinya dengan SEM dan uji pelepasan klindamisin menggunakan spektrofotometri UV-Vis. Hasil XRD menunjukkan terbentuknya fasa HAp dengan kristalinitas 95,42%, meskipun terdeteksi fasa residu kalsium oksida akibat sintesis yang belum sepenuhnya optimal. Karakterisasi SEM menunjukkan MNs berbentuk kerucut dengan distribusi seragam, lebar basis sekitar 530 µm, tinggi sekitar 600 µm, dan jarak antar jarum sekitar 600 µm, mendukung potensi penetrasi transdermal yang efektif. Pengujian UV-Vis mengonfirmasi pelepasan klindamisin dengan serapan maksimum pada panjang gelombang 232,5 nm, meskipun ditemukan puncak tambahan yang mengindikasikan kontribusi zat lain dalam media pelepasan.
       
      Bacterial skin infections caused by antibiotic-resistant Staphylococcus aureus present a challenge that requires more effective drug delivery systems than conventional oral and topical methods. This study aims to determine the physical characteristics of hydroxyapatite (HAp)-based microneedles (MNs) and the release of clindamycin within the delivery system across formulation variations. HAp was synthesized from chicken eggshells through calcination and reaction with phosphoric acid, then characterized using XRD. A Gel/HAp composite mixed with clindamycin and glutaraldehyde was molded into MNs using a PDMS mold, followed by morphological characterization with SEM and clindamycin release testing using UV-Vis spectrophotometry. XRD results showed the formation of an HAp phase with 95,42% of crystallinity, although a residual calcium oxide phase was detected due to incomplete synthesis optimization. SEM characterization revealed cone-shaped MNs with uniform distribution, a base width of approximately 530 µm, a height of approximately 600 µm, and an inter-needle spacing of approximately 600 µm, supporting effective transdermal penetration potential. UV-Vis testing confirmed clindamycin release with maximum absorbance at a wavelength of 232,5 nm, although additional peaks indicated contributions from other substances in the release medium.
