      Abstract
      Acne vulgaris merupakan salah satu penyakit kulit inflamasi kompleks dengan prevalensi tinggi secara global, sementara tata laksana terapinya saat ini masih didominasi penggunaan antibiotik di tengah meningkatnya ancaman resistansi antimikroba. Imunoglobulin Y (IgY) merupakan alternatif terapi non-antibiotik yang lebih aman dan ekonomis. Penelitian ini bertujuan menghasilkan dan mengarakterisasi antibodi IgY trivalen terhadap tiga bakteri utama penyebab acne vulgaris, yaitu Staphylococcus aureus (SA), Staphylococcus epidermidis (SE), dan Cutibacterium acnes (CA), sebagai dasar pengembangan imunoterapi topikal anti-acne. Sebanyak lima ekor ayam petelur diimunisasi menggunakan vaksin bakteri utuh inaktif trivalen. Respons antibodi dalam serum dan kuning telur diamati selama 12 minggu menggunakan metode Agar Gel Precipitation Test (AGPT). Isolasi IgY dilakukan melalui metode water-soluble fraction (WSF) yang dilanjutkan dengan presipitasi NaCl 8,8% (b/v), kemudian dibandingkan dengan kit komersial. Antibodi hasil purifikasi dikarakterisasi menggunakan uji Bradford, SDS-PAGE, dan AGPT fungsional. Hasil penelitian menunjukkan bahwa titer total IgY pada kuning telur relatif stabil akibat integrasi temporal selama proses vitelogenesis, sedangkan titer serum berfluktuasi. Kinetika antibodi mengindikasikan adanya kompetisi antigen yang ditunjukkan oleh fenomena imunodominansi terhadap SA dengan titer tinggi yang bertahan hingga minggu ke-12 (±6 log2). Sebaliknya, titer antibodi terhadap SE dan CA mengalami penurunan setelah minggu ke-6. Metode WSF-NaCl menghasilkan antibodi murni dengan konsentrasi protein sebesar 1,150 mg/mL. Analisis struktur menggunakan SDS-PAGE mengonfirmasi integritas IgY melalui kemunculan pita rantai berat (65-70 kDa) dan rantai ringan (25-27 kDa). Uji AGPT fungsional menunjukkan bahwa IgY hasil purifikasi memiliki spesifisitas terhadap antigen trivalen yang digunakan.
       
      Acne vulgaris is one of the most prevalent complex inflammatory skin disorders, and current therapies still rely on antibiotics, despite the increasing burden of antimicrobial resistance. Immunoglobulin Y (IgY) presents as a safer and more economical non-antibiotic alternative. This study aimed to produce and characterize trivalent IgY antibodies against three major acne-associated pathogens: Staphylococcus aureus (SA), Staphylococcus epidermidis (SE), and Cutibacterium acnes (CA), as a basis for the topical anti-acne immunotherapy. Five laying hens were immunized with a trivalent inactivated whole-cell bacterial vaccine. Antibody responses in serum and egg yolk were monitored for 12 weeks by Agar Gel Precipitation Test (AGPT). IgY was isolated via the water-soluble fraction (WSF) method, followed by 8.8% (w/v) NaCl precipitation, then the result was compared against a commercial kit. Purified antibody was characterized using Bradford assay, SDS-PAGE, and functional AGPT. The result demonstrated that total IgY titers in yolk remained stable due to the temporal integration of the vitellogenesis process, while the serum titers showed fluctuations. Antibody kinetics indicated an antigenic competition manifested as immunodominance by SA, with sustained high titers (~6 log2) through week 12. In contrast, antibody titers against SE and CA declined following week 6. The WSF-NaCl method yielded a purified antibody with a protein concentration of 1.150 mg/mL. Structural analysis via SDS-PAGE confirmed the integrity of IgY, with heavy chain (65-70 kDa) and light chain (25-27 kDa) bands. Functional AGPT confirmed that the purified IgY successfully shows specificity against the trivalent antigen.
