      Abstract
      Peniophora sp. merupakan cendawan pelapuk kayu yang memiliki potensi patogenik pada tanaman berkayu. Penelitian ini bertujuan mengevaluasi potensi cendawan Polyporales dan endofit asal tanaman pingku (Dysoxylum parasiticum) serta buah naga sebagai agens hayati terhadap Peniophora sp. in vitro, sekaligus menilai kemampuan pemacu pertumbuhan tanaman dan keamanan hayatinya. Identifikasi molekuler patogen dilakukan menggunakan primer ITS1–ITS4 yang dilanjutkan dengan analisis filogenetik. Aktivitas antagonistik dievaluasi melalui uji interaksi, antibiosis, dan produksi senyawa organik volatil, serta pengujian produksi enzim glukanolitik dan kitinolitik, produksi indole-3-acetic acid (IAA), pelarutan fosfat, aktivitas hemolisis, dan uji patogenisitas pada padi. Sebanyak 24 isolat menunjukkan aktivitas antagonistik yang didominasi oleh mekanisme antibiosis. Isolat SKLH menunjukkan penghambatan tertinggi pada uji dual culture sebesar 57%, sedangkan isolat BWC6 menunjukkan penghambatan tertinggi pada uji VOC sebesar 62%. Delapan isolat menunjukkan aktivitas glukanolitik dan 10 isolat mampu memproduksi IAA, sementara aktivitas kitinolitik tidak terdeteksi. Dua cendawan Polyporales dengan kode PLY 1 dan PLY 2 menunjukkan hasil positif hemolisis, sedangkan lima isolat bersifat non-patogenik pada uji patogenisitas padi. Hasil penelitian menunjukkan bahwa isolat terpilih berpotensi sebagai agens hayati yang efektif dan relatif aman.
       
      Peniophora sp. is a wood-decaying fungus with potential pathogenicity on woody plants and remains challenging to manage using environmentally friendly approaches. This study aimed to evaluate Polyporales fungi and endophytes from pingku tree (Dysoxylum parasiticum) and dragon fruit as biological control agents against Peniophora sp. in vitro, and to assess their plant growth-promoting ability and biosafety. Molecular identification used ITS1–ITS4 primers targeting ITS DNA, followed by phylogenetic analysis. Antagonistic activity was evaluated through interaction test, antibiosis, and volatile organic compound (VOC) assays, together with glucanolytic and chitinolytic enzyme production, indole-3-acetic acid production, phosphate solubilization, hemolysis, and rice pathogenicity tests. Twenty-four isolates exhibited antagonistic activity, predominantly through antibiosis. SKLH showed the highest dual-culture inhibition at 57%, whereas BWC6 showed the highest VOC inhibition at 62%. Eight isolates showed glucanolytic activity and 10 produced IAA, while chitinolytic activity was not detected. Two Polyporales fungi, PLY 1 and PLY 2, showed positive hemolysis, while five isolates were non-pathogenic in the rice pathogenicity test. These findings indicate that the selected isolates have strong potential as effective and safe biological control agents.
