dc.description.abstractEscherichia coli (E. coli) merupakan bakteri indikator resistansi antibiotik yang banyak ditemukan pada kotoran sapi perah dan berperan sebagai reservoir penyebaran gen resistansi antibiotik di lingkungan peternakan. Penelitian ini bertujuan mendeteksi dan menganalisis gen resistansi antibiotik golongan betalaktam (bla) pada E. coli yang diisolasi dari kotoran sapi perah di Kota Bogor. Sebanyak 15 isolat E. coli dari 15 peternakan sapi perah di Kebon Pedes yang telah dikonfirmasi pada penelitian sebelumnya digunakan dalam penelitian ini. Gen resistansi beta-laktam (bla) pada isolat E. coli dideteksi menggunakan teknologi MinION Oxford Nanopore Technologies, kemudian dianalisis secara bioinformatika untuk mengidentifikasi profil gen resistansi antibiotik. Seluruh isolat E. coli (100%) membawa gen resistansi antibiotik beta-laktam dengan distribusi satu hingga lebih dari empat gen resistansi. Gen yang paling dominan adalah blaCTX-M-55 dan blaTEM-1B sebesar 53,33%, diikuti blaOXA-10 sebesar 40,00%. Sebagian besar gen resistansi berada pada plasmid (97,30%), sedangkan 2,70% teridentifikasi pada kromosom. Hasil penelitian ini menunjukkan tingginya potensi penyebaran gen resistansi antibiotik beta-laktam di lingkungan peternakan sapi perah.
dc.description.abstractEscherichia coli (E. coli) is an important indicator of antibiotic resistance and is commonly found in dairy cattle manure, where it serves as a reservoir for the spread of resistance genes in farm environments. This study aimed to detect and characterize beta-lactam resistance genes (bla) in E. coli isolated from dairy cattle manure in Bogor, Indonesia. A total of 15 Escherichia coli (E. coli) isolates obtained from 15 dairy farms in Kebon Pedes and previously confirmed in earlier studies were included in this study. Beta-lactam resistance genes (bla) in E. coli isolates were detected using MinION Oxford Nanopore Technologies, followed by bioinformatic analysis to characterize antibiotic resistance gene profiles. The dominant genes identified were blaCTX-M-55 and blaTEM-1B, each detected in 53.33% of the isolates, followed by blaOXA-10 40.00%. Most resistance genes were located on plasmids 97.30%, whereas only 2.70% were identified on chromosomes. These findings indicate a high potential for the spread of beta-lactam resistance genes through horizontal gene transfer in dairy farm environments.
