      Kandungan Metabolit Sekunder Arachis pintoi dan Potensinya sebagai Bioherbisida untuk Pengendalian Gulma Tanaman Padi (Oryza sativa L.)

      Abstract
      Gulma menjadi salah satu faktor pembatas produksi padi karena keberadaannya menyebabkan kompetisi dengan tanaman budidaya dalam pemanfaatan cahaya, air, unsur hara, dan ruang tumbuh. Penggunaan herbisida sintetis secara berkelanjutan dan dalam jumlah tinggi berpotensi menimbulkan dampak negatif terhadap lingkungan, sehingga diperlukan alternatif pengendalian gulma yang lebih ramah lingkungan. Tanaman Arachis pintoi diketahui mengandung berbagai senyawa metabolit sekunder yang berpotensi dimanfaatkan sebagai bioherbisida. Penelitian ini bertujuan untuk mengidentifikasi profil metabolit sekunder ekstrak Arachis pintoi serta mengevaluasi efektivitasnya sebagai bioherbisida pre-emergence, early post-emergence, dan post-emergence terhadap beberapa jenis gulma utama pada pertanaman padi. Penelitian menggunakan rancangan acak lengkap dengan satu faktor yaitu konsentrasi ekstrak Arachis pintoi yang terdiri atas 0, 50, 100, 150, 200, dan 250 g/L. Identifikasi metabolit sekunder dilakukan menggunakan metode Liquid Chromatography–Mass Spectrometry (LC–MS), sedangkan pengujian aktivitas bioherbisida dilakukan terhadap Ludwigia octovalvis, Cyperus iria, Eleusine indica, dan padi (Oryza sativa L.). Hasil analisis LC–MS menunjukkan bahwa ekstrak Arachis pintoi mengandung 15 senyawa metabolit sekunder yang termasuk dalam kelompok asam alkaloid, flavonoid, fenolik, serta terpenoid yang berpotensi berperan sebagai alelokimia. Pada fase pre-emergence, ekstrak Arachis pintoi secara nyata menghambat perkecambahan dan pertumbuhan awal kecambah seluruh tanaman uji dengan tingkat inhibisi tertinggi pada Ludwigia octovalvis yang mencapai 100% pada konsentrasi = 200 g/L. Pada fase early post-emergence, ekstrak Arachis pintoi masih mampu menekan persentase kemunculan kecambah dan bobot kering Ludwigia octovalvis. Fase post-emergence, ekstrak Arachis pintoi menurunkan jumlah daun dan bobot kering Ludwigia octovalvis serta meningkatkan gejala fitotoksisitas pada Eleusine indica, tetapi tidak berpengaruh nyata terhadap pertumbuhan padi. Berdasarkan hasil penelitian, ekstrak Arachis pintoi berpotensi dikembangkan sebagai bioherbisida, khususnya untuk pengendalian Ludwigia octovalvis pada pertanaman padi.
       
      Weeds are one of the limiting factors in rice production because their presence causes competition with cultivated plants for light, water, nutrients, and growing space. The continuous use of synthetic herbicides in high quantities has the potential to cause negative environmental impacts, making it necessary to find more environmentally friendly alternatives for weed control. The Arachis pintoi plant is known to contain various secondary metabolites that have the potential to be utilized as bioherbicides. This study aims to identify the secondary metabolite profile of Arachis pintoi extract and evaluate its effectiveness as a pre-emergence, early post-emergence, and post-emergence bioherbicide against several major weed species in rice fields. The study employed a completely randomized design with a single factor, Arachis pintoi extract concentration consisting of 0, 50, 100, 150, 200, and 250 g/L. Secondary metabolites were identified using Liquid Chromatography Mass Spectrometry (LC–MS), while bioherbicide activity testing was conducted on Ludwigia octovalvis, Cyperus iria, Eleusine indica, and rice (Oryza sativa L.). LC-MS analysis results showed that Arachis pintoi extract contained 15 secondary metabolites belonging to the groups of alkaloids, flavonoids, phenolics, and terpenoids that may act as allelochemicals. In the pre-emergence, Arachis pintoi extract significantly inhibited germination and early seedling growth in all test plants, with the highest inhibition rate observed in Ludwigia octovalvis, reaching 100% at concentrations = 200 g/L. In the early post-emergence, Arachis pintoi extract continued to suppress the germination rate and dry weight of Ludwigia octovalvis. In the post-emergence, Arachis pintoi extract reduced the number of leaves and dry weight of Ludwigia octovalvis and increased phytotoxicity symptoms in Eleusine indica, but had no significant effect on rice growth. Based on the research results, Arachis pintoi extract has the potential to be developed as a bioherbicide, particularly for the control of Ludwigia octovalvis in rice fields.
