| dc.description.abstract | Aphelenchoides fragariae merupakan nematoda OPTK A2 yang dapat berperan sebagai fungivora. Perilaku tersebut dapat dimanfaatkan untuk perbanyakan secara in vitro menggunakan media cendawan. Penelitian ini bertujuan mengevaluasi kesesuaian perkembangan A. fragariae pada cendawan endofit dan patogen serta mengkaji keterkaitan kandungan nutrisinya. Cendawan yang digunakan meliputi Alternaria porri, Rhizopus sp., Cladosporium sp., dan Talaromyces verruculosus. Sebanyak 40 nematoda diinfestasikan pada setiap biakan cendawan kemudian diinkubasi selama 21 hari pada suhu 28 °C untuk dihitung populasi akhirnya. Hasil penelitian menunjukkan A. porri merupakan media perbanyakan paling sesuai dengan rata-rata populasi akhir tertinggi hingga 708 nematoda, sebaliknya T. verruculosus tidak mampu mendukung perkembangan nematoda. Hasil analisis asam amino menggunakan HPLC menunjukkan total kadar asam amino pada T. verruculosus (14.400 mg/kg) lebih tinggi dibandingkan A. porri (1.166,10 mg/kg). Hasil penelitian membuktikan bahwa kadar asam amino pada cendawan tidak berbanding lurus dengan faktor reproduksi A. fragariae. Asam amino bukan faktor utama dalam menentukan keberhasilan reproduksi nematoda. Penelitian ini menunjukkan bahwa A. porri berpotensi sebagai media perbanyakan massal A. fragariae. | |
| dc.description.abstract | Aphelenchoides fragariae is an A2 quarantine pest nematode that can also function as a fungivore. This behavior can be utilized for its in vitro propagation using fungal culture media. This study evaluated the suitability of endophytic and pathogenic fungi for the development of A. fragariae and examined their relationship with fungal nutritional content. The fungi tested were Alternaria porri, Rhizopus sp., Cladosporium sp., and Talaromyces verruculosus. Forty nematodes were inoculated onto each fungal culture and incubated at 28 °C for 21 days, after which the final population was assessed. The results showed that A. porri was the most suitable propagation medium, supporting the highest average final population of 708 nematodes, whereas T. verruculosus failed to support nematode development. Amino acid analysis using HPLC showed that the total amino acid content of T. verruculosus (14.400 mg/kg) was higher than that of A. porri (1.166,10 mg/kg). These findings demonstrate that the amino acid content of fungal cultures is not directly correlated with the reproductive factor of A. fragariae, indicating that amino acids are not the primary determinant of successful nematode reproduction. These findings indicate that A. porri has potential as a medium for the mass propagation of A. fragariae. | |
