      Identifikasi Molekuler Tungau Laba-laba Merah (Acari: Tetranychidae) pada Tanaman Ubi Kayu dan Evaluasi Kemampuan Kitosan untuk Mengendalikannya

      Abstract
      Tungau laba-laba merah merupakan hama penting pada tanaman ubi kayu yang berpotensi menyebabkan kerusakan daun dan penurunan hasil. Penelitian ini bertujuan mengidentifikasi secara molekuler tungau laba-laba merah yang menyerang tanaman ubi kayu dan mengevaluasi kemampuan larutan kitosan dan nanopartikel kitosan dalam mengendalikan populasi tungau laba-laba merah. Tungau dikoleksi dari pertanaman ubi kayu di BRMP Cipanas, Jawa Barat. Evaluasi kemampuan kitosan non nanopartikel dalam mengendalikan tungau dilakukan dengan penyemprotan langsung pada konsentrasi 500, 800, dan 1.000 ppm, dan nanopartikel kitosan pada konsentrasi 455, 727, dan 909 ppm. Akuabides dan akarisida berbahan aktif piridaben digunakan sebagai membanding. Mortalitas diamati pada 24, 48, dan 72 jam setelah perlakuan. PCR tungau laba-laba merah pada ubi kayu dengan primer universal rDNA Tetranychidae rD02 dan HC2 teramplifikasi DNA berukuran ± 1.100 bp. Berdasarkan runutan rDNA, tungau laba-laba merah pada ubi kayu teridentifikassi sebagai Tetranychus kanzawai, dengan similaritas tertinggi sebesar 99,90% dengan T. kanzawai Khisida asal Jepang dan Taiwan. Perlakuan kitosan non nanopartikel pada konsentrasi 1.000 ppm mampu menyebabkan mortalitas tungau laba-laba merah tertinggi sebesar 86,67%, dibandingkan dengan nanopartikel kitosan sebesar 12,50%. Hal ini mengindikasikan potensi kitosan dalam mengendalikan tungau pada ubi kayu.
       
      The red spider mite is an important pest of cassava that can cause severe leaf damage and yield reduction. This study aimed to molecularly identify the red spider mites infesting cassava and to evaluate the efficacy of chitosan solution and chitosan nanoparticles in controlling its population. Mites were collected from cassava fields BRMP, Cipanas, West Java. The efficacy of non-nanoparticle chitosan was evaluated by direct spraying at concentrations of 500, 800, and 1,000 ppm, whereas chitosan nanoparticles were applied at concentrations of 455, 727, and 909 ppm. Sterile distilled water and the acaricide containing pyridaben as the active ingredient were used as the negative and positive controls. Mortality was recorded at 24, 48, and 72 hours after treatment. PCR amplification of the red spider mite infesting cassava using the universal rDNA Tetranychidae primers rD02 and HC2 produced a DNA fragment of approximately 1,100 bp. Based on the rDNA sequence, the red spider mite infesting cassava was identified as Tetranychus kanzawai, showing the highest sequence similarity 99.90% to T. kanzawai Kishida from Japan and Taiwan. Non nanoparticle chitosan at a concentration of 1,000 ppm resulted in the highest mortality of the red spider mites 86.67%, compared with chitosan nanoparticles 12.50%. These findings indicate the potential of chitosan for controlling red spider mites on cassava.
