      Abstract
      Kultur jaringan merupakan pendekatan produksi bibit rumput laut kotoni berkualitas tinggi secara berkelanjutan, khususnya dalam mengatasi tantangan yang terkait dengan variabilitas iklim dan wabah penyakit. Penelitian ini bertujuan untuk mengevaluasi pengaruh penambahan Indole Acetic Acid (IAA) terhadap regenerasi mikropropagul dan menentukan konsentrasi IAA optimal. Metode yang digunakan adalah perlakuan media Provasoli’s Enriched Seawater (PES) dengan konsentrasi 20 mL L-1 ditambahkan konsentrasi ZPT IAA dengan Rancangan Acak Lengkap (RAL) empat perlakuan dan lima ulangan, yaitu IK (kontrol), I1 (0,1 mg L-1 IAA), I2 (0,2 mg L-1 IAA), dan I3 (0,3 mg L-1 IAA). Hasil penelitian menunjukkan bahwa perlakuan I1 menghasilkan jumlah mikropropagul tertinggi, mencapai 258,00±7,41 individu pada akhir periode induksi. Seluruh perlakuan mempertahankan persentase kelangsungan hidup dan pertumbuhan eksplan yang tinggi selama periode kultur. Penambahan 0,1 mg L-1 IAA merupakan konsentrasi paling efektif untuk mendorong regenerasi mikropropagul.
       
      Tissue culture is an approach for ensuring the sustainable production of high-quality cottonii seaweed seedlings, particularly in overcoming challenges associated with climate variability and disease outbreaks. This study aimed to evaluate the effects of Indole Acetic Acid (IAA) supplementation on micropropagule regeneration and to determine the optimal IAA concentration. The method employed Provasoli’s Enriched Seawater (PES) medium at a concentration of 20 mL L?¹ supplemented with different concentrations of IAA in a Completely Randomized Design (CRD) with four treatments and five replicates: IK (control), I1 (0,1 mg L?¹ IAA), I2 (0,2 mg L?¹ IAA), and I3 (0,3 mg L?¹ IAA). The results demonstrated that the I1 treatment produced the highest number of micropropagules, reaching 258.00±7.41 individuals at the end of the induction period. All treatments maintained high explant survival and growth percentages throughout the culture period. Supplementation with 0,1 mg L-1 IAA is the most effective concentration for promoting micropropagule regeneration.
