dc.description.abstractNematoda gastrointestinal merupakan parasit penting pada ruminansia liar dan identifikasinya sering terkendala oleh kemiripan morfologi antarspesies pada stadium larva. Penelitian ini bertujuan mengidentifikasi nematoda gastrointestinal pada rusa timor (Cervus timorensis) di Taman Satwa Tangerang Selatan melalui analisis morfometrik larva stadium tiga (L3) dan analisis filogenetik. Larva L3 diperoleh melalui kultur feses, diamati secara morfometrik, dan dianalisis secara molekuler melalui PCR, sekuensing, BLAST, serta konstruksi pohon filogenetik dengan metode Neighbor-Joining. Hasil elektroforesis menunjukkan pita DNA pada kisaran 300–400 bp. Setelah trimming, sekuens konsensus memiliki panjang 239 bp. Hasil BLAST menunjukkan query cover 100%, percent identity 98,74% dengan Spiculopteragia houdemeri, dan nilai E-value 1e-114. Analisis filogenetik menempatkan isolat penelitian dalam satu klad dengan S. Houdemeri. Hasil ini menunjukkan bahwa kombinasi analisis morfometrik dan ITS2 efektif untuk identifikasi nematoda gastrointestinal secara akurat.
dc.description.abstractGastrointestinal nematodes are important parasites of wild ruminants, and species identification is often constrained by morphological similarity among larval stages. This study aimed to identify gastrointestinal nematodes infecting Timor deer (Cervus timorensis) at Tangerang Selatan Zoo using morphometric analysis of third-stage larvae (L3) and phylogenetic analysis. L3 larvae were obtained through fecal culture, examined morphometrically, and analyzed molecularly through PCR, sequencing, BLAST analysis, and phylogenetic tree construction using the Neighbor-Joining method. Gel electrophoresis showed a PCR product in the 300–400 bp range. After trimming, the consensus sequence length was 239 bp. BLAST analysis showed 100% query coverage, 98.74% similarity to Spiculopteragia houdemeri, and an E-value of 1e-114. Phylogenetic analysis placed the isolate in the same clade as S. houdemeri. These findings indicate that combining morphometric and ITS2-based molecular analyses is effective for accurate identification of gastrointestinal nematodes.
