      PERBANDINGAN PENGARUH ß-HYDROXYETHYL PHENYL ETHER DAN TRICAINE METHANESULFONATE TERHADAP PROFIL DARAH PADA IKAN KOI (Cyprinus carpio haematopterus)

      Abstract
      Pemeriksaan darah merupakan salah satu metode penting dalam manajemen kesehatan koi (Cyprinus carpio haematopterus). Namun dalam proses pengambilan darah sering kali menimbulkan stres sehingga diperlukan agen anestesi pada tahap sedasi untuk meminimalkan perubahan fisiologis selama proses pengambilan darah. Penelitian ini bertujuan mengevaluasi pengaruh ß-hydroxyethyl phenyl ether atau 2-phenoxyethanol (2-PE) dibandingkan dengan tricaine methanesulfonate (MS-222) terhadap hematologi, kimia darah, dan blood performance pada ikan koi. Sebanyak 15 ekor ikan koi dibagi menjadi tiga kelompok: kontrol negatif (tanpa sedasi), kelompok yang diberi 2-PE (0,2 mL/L), dan kelompok yang diberi tricaine methanesulfonate (MS-222) (50 mg/L) sebagai pembanding standar. Parameter hematologi dianalisis menggunakan metode standar dan kimia darah dianalisis menggunakan mesin otomatis kimia darah. Data dianalisis menggunakan uji ANOVA atau Kruskal-Wallis pada tingkat kepercayaan 95%. Hasil penelitian menunjukkan bahwa pemberian sedasi tidak memberikan pengaruh nyata (p>0,05) terhadap sebagian besar parameter hematologi, kimia darah, maupun indeks blood performance. Perbedaan nyata (p<0,05) hanya ditemukan pada variabel ion fosfat yang lebih rendah pada kelompok sedasi dibandingkan dengan kelompok kontrol. Hasil tersebut menunjukkan bahwa penggunaan 2-PE pada tahap sedasi tidak menunjukkan perbedaan yang signifikan pada sebagian besar parameter darah yang diamati dibandingkan kelompok kontrol maupun MS-222 sehingga berpotensi digunakan sebagai agen sedasi pada prosedur pengambilan sampel darah ikan koi.
       
      Blood examination is an important component of health management in koi fish (Cyprinus carpio haematopterus). However, blood sampling procedures may induce stress, requiring anesthetic agents at the sedation stage to maintain physiological stability. This study aimed to evaluate the effects of ß-hydroxyethyl phenyl ether or 2-phenoxyethanol (2-PE), in comparison with tricaine methanesulfonate (MS-222), on hematological, blood biochemical, and blood performance parameters in koi fish. Fifteen koi fish were assigned to three groups: a negative control (without sedation), a 2-PE group (0.2 mL/L), and an MS-222 group (50 mg/L) as the comparator group. Hematological parameters were analyzed using standard methods, whereas blood biochemical parameters were measured using an automatic blood chemistry analyzer. Data were analyzed using ANOVA or the Kruskal–Wallis test at a 95% confidence level. The results showed no significant differences (p>0.05) in most hematological, blood biochemical, or blood performance parameters. A significant difference (p<0.05) was observed only in phosphate ion levels which were significantly higher in the control group than in the sedation groups. These findings indicate that 2-PE at the sedation stage did not produce statistically significant differences in most measured blood parameters compared with the control and MS-222 groups.
