      Pembedaan Hemosit Ulat Sutra Liar Attacus atlas (Lepidoptera: Saturniidae) pada Suhu dan Waktu Penyimpanan Berbeda

      Abstract
      Attacus atlas atau kupu-kupu gajah merupakan ulat sutra liar penghasil kokon yang berpotensi dibudidaya karena menghasilkan serat sutra yang bersifat antibakteri, tahan panas, dan antiultraviolet. Hemolimfa merupakan cairan utama serangga yang berperan dalam perpindahan nutrisi dan menjaga keseimbangan fisiologik, sedangkan hemosit sebagai bagian sel berfungsi dalam reaksi pertahanan tubuh bawaan yang jumlahnya berubah selama metamorfosis. Penelitian bertujuan menganalisis perubahan differential hemocyte count (DHC) larva tahap akhir instar VI A. atlas setelah penyimpanan pada suhu ruang 26 °C dan suhu kulkas 4 °C selama 0, 7, 14, dan 28 hari. Hemolimfa diambil dengan memotong kaki depan larva dan dibuat preparat ulas darah dengan pewarnaan Giemsa 10%. Pengamatan DHC menggunakan mikroskop objektif perbesaran 100× dan analisis statistik two way ANOVA. Hasil pengamatan menunjukkan lima tipe hemosit, yaitu prohemosit, plasmatosit, granulosit, sferulosit, dan oenositoid. Selama penyimpanan terjadi perubahan DHC yang dipengaruhi waktu dan suhu penyimpanan. Waktu berpengaruh nyata (P<0,05) pada perubahan DHC, suhu tidak berpengaruh nyata (P>0,05), interaksi suhu dan waktu berpengaruh nyata (P<0,05) pada plasmatosit. Prohemosit menurun hingga hari ke-14 kemudian meningkat pada hari ke-28. Plasmatosit berbanding terbalik dengan granulosit, sedangkan sferulosit dan oenositoid menurun hingga akhir pengamatan. Simpulan penelitian ini, penyimpanan suhu kulkas memperlambat perubahan fisiologik dan metabolisme dibanding suhu ruang.
       
      Attacus atlas (Atlas moth) is a wild silkworm that produces cocoons with potential for commercial cultivation because its silk fibers possess antibacterial, heat-resistant, and ultraviolet-protective properties. Hemolymph is the primary circulatory fluid in insects, functioning in nutrient transport and physiological homeostasis, while hemocytes are responsible for innate immune responses and undergo changes during metamorphosis. This study aimed to analyze changes in the differential hemocyte count (DHC) of final sixth-instar A. atlas larvae after hemolymph storage at room temperature (26 °C) and refrigeration temperature (4 °C) for 0, 7, 14, and 28 days. Hemolymph was collected by severing the larval foreleg, and hemolymph smears stained with 10% Giemsa were prepared for DHC observation under a light microscope (100× objective). Data were analyzed using two-way ANOVA. Five hemocyte types were identified: prohemocytes, plasmatocytes, granulocytes, spherulocytes, and oenocytoids. DHC changed during storage and was influenced by storage duration and temperature. Storage duration significantly affected DHC (P<0.05), whereas storage temperature had no significant effect (P>0.05). A significant interaction between storage duration and temperature was observed only for plasmatocytes (P<0.05). Refrigerated storage slowed physiological and metabolic changes compared with room-temperature storage.
