      Optimasi Metode Ekstraksi DNA dari Telur Trematoda

      Abstract
      Telur trematoda memiliki dinding yang tebal dan kompleks sehingga menjadi tantangan dalam proses ekstraksi DNA. Kondisi ini dapat memengaruhi keberhasilan analisis molekuler. Penelitian ini bertujuan mengoptimasi metode ekstraksi DNA telur trematoda melalui penambahan tahap preparasi berupa vibrasi mekanik pada sampel pool tiga telur. Ekstraksi DNA dilakukan menggunakan lysis solution dengan dan tanpa perlakuan vibrasi mekanik, kemudian dievaluasi berdasarkan konsentrasi DNA menggunakan NanoDrop dan visualisasi hasil amplifikasi Polymerase Chain Reaction (PCR) melalui elektroforesis. Hasil analisis NanoDrop menunjukkan bahwa perlakuan vibrasi menghasilkan konsentrasi DNA lebih tinggi (355 ng/µL) dibandingkan lysis solution saja (199,6 ng/µL). Kedua perlakuan menghasilkan pita DNA yang jelas berukuran 400 bp yang relatif tidak berbeda. Penggunaan lysis solution dengan volume reaksi PCR 10 µL dinilai optimal untuk menghasilkan DNA yang dapat diamplifikasi, tanpa memerlukan tambahan vibrasi mekanik.
       
      Trematode eggs possess thick and complex eggshells, making DNA extraction challenging. This condition may affect the success of molecular analyses. This study aimed to optimize the DNA extraction method for trematode eggs by incorporating a mechanical vibration pretreatment into pooled samples containing three eggs. DNA extraction was performed using a lysis solution with and without mechanical vibration pretreatment, followed by evaluation of DNA concentration using a NanoDrop spectrophotometer and visualization of Polymerase Chain Reaction (PCR) amplification products by agarose gel electrophoresis. NanoDrop analysis showed that the vibration treatment yielded a higher DNA concentration (355 ng/µL) than the lysis solution alone (199.6 ng/µL). Both treatments produced distinct DNA bands of approximately 400 bp with relatively similar band intensities. The use of the lysis solution with a 10 µL PCR reaction volume was considered optimal for obtaining amplifiable DNA without requiring additional mechanical vibration.
