      Analisis Morfometrik dan Molekuler Nematoda Parasit Trichostrongylus sp. Pada Domba

      Abstract
      Infeksi Trichostrongylus sp. merupakan salah satu penyebab menurunnya produktivitas domba, tetapi informasi mengenai karakteristiknya di Indonesia masih terbatas sehingga diperlukan identifikasi yang lebih akurat. Penelitian ini bertujuan untuk menganalisis karakter morfometrik dan molekuler Trichostrongylus sp. pada domba serta membandingkan kesesuaian hasil identifikasi dari kedua pendekatan tersebut. Sampel feses domba dikultur untuk memperoleh larva infektif (L3), kemudian dianalisis secara morfometrik melalui pengamatan mikroskopis dan secara molekuler melalui amplifikasi fragmen ITS-2, sekuensing, BLAST, serta konstruksi pohon filogenetik. Larva menunjukkan karakter morfologi khas genus Trichostrongylus dengan panjang tubuh sekitar 702 µm. Sekuens ITS-2 memiliki kemiripan 99,21% dengan Trichostrongylus axei dan menempatkan isolat Bogor dalam satu klad bersama isolat T. axei dari berbagai negara. Berdasarkan kesesuaian hasil morfometrik dan molekuler, nematoda gastrointestinal pada domba dalam penelitian ini teridentifikasi sebagai Trichostrongylus axei.
       
      Infection with Trichostrongylus sp. is a major constraint to sheep productivity. However, information on its characteristics in Indonesia remains limited, necessitating more accurate identification. This study aimed to analyze the morphometric and molecular characteristics of Trichostrongylus sp. in sheep and to compare the consistency of identification results between the two approaches. Sheep fecal samples were cultured to obtain infective larvae (L3), which were then analyzed morphometrically by microscopy and molecularly by amplification of the ITS-2 fragment, sequencing, BLAST analysis, and phylogenetic tree construction. The larvae exhibited morphological characteristics typical of the genus Trichostrongylus with a body length of approximately 702 µm. The ITS-2 sequence showed 99.21% similarity to Trichostrongylus axei and placed the Bogor isolate within the same clade as T. axei isolates from various countries. Based on the agreement between morphometric and molecular results, the gastrointestinal nematode in sheep in this study was identified asTrichostrongylus axei.
