Studi Pendahuluan Identifikasi Morfologi dan Analisis Filogenetik Cyathostominae pada Kuda (Equus caballus)

Abstract

Cyathostominae merupakan nematoda gastrointestinal yang dominan pada kuda. Identifikasi secara morfologi terhambat oleh kemiripan antarspesies yang tinggi. Penelitian ini bertujuan mengidentifikasi karakteristik morfologi larva stadium ketiga (L3) dan menentukan posisi taksonomi isolat asal Bogor melalui pendekatan morfologi dan molekuler. Kultur feses seekor kuda dari Unit Rehabilitasi Reproduksi SKHB IPB berhasil mengisolasi larva L3 dengan ciri khas delapan sel usus abdomen, dengan rata-rata ukuran panjang total 648,59±29,74 µm dan tubuh 20,24±0,05 µm. Analisis molekuler pada regio Internal Transcribed Spacer 2 (ITS-2) berhasil mengamplifikasi fragmen DNA sepanjang 278 bp. Hasil analisis Basic Local Alignment Search Tool (BLAST) mengonfirmasi persentase kemiripan sebesar 100% dengan query coverage 90% pada Coronocyclus coronatus asal Jerman (KY747448). Konstruksi pohon filogenetik memperkuat hasil tersebut dan menempatkan isolat Bogor dalam klad monofiletik bersama C. coronatus pada jarak genetik 0,000. Dapat disimpulkan bahwa Cyathostominae pada kuda asal Bogor adalah C. coronatus.

Cyathostominae are the dominant gastrointestinal nematodes in horses. High interspecies similarity hinders morphological identification. This study aims to identify the morphological characteristics of third-stage larvae (L3) and determine the taxonomic position of the Bogor isolate through morphological and molecular approaches. Fecal culture from a horse at the Rehabilitation and Reproduction Unit of SKHB IPB successfully isolated ??3 larvae characterized by eight abdominal intestinal cells, an average total length of 648.59±29.74 µm, and a body of 20.24±0.05 µm. Molecular analysis of the Internal Transcribed Spacer 2 (ITS-2) region successfully amplified a 278 bp DNA fragment. The Basic Local Alignment Search Tool (BLAST) analysis results confirmed a 100% similarity percentage with 90% query coverage to Coronocyclus coronatus from Germany (KY747448). Phylogenetic tree construction reinforced these results, placing the Bogor isolate within a monophyletic clade alongside C. coronatus at a genetic distance of 0.000. This study concludes that the Cyathostominae found in horses from Bogor is C. coronatus.
