      Abstract
      Mastitis subklinis merupakan salah satu penyakit penting pada sapi perah yang menyebabkan kerugian ekonomi akibat penurunan produksi dan kualitas susu. Salah satu agen penyebab utama penyakit ini adalah Streptococcus agalactiae. Pengendalian mastitis umumnya dilakukan melalui penggunaan antibiotik, namun pemakaian berulang berisiko menimbulkan residu dan resistensi antimikroba. Oleh karena itu, pengembangan vaksin menjadi salah satu alternatif strategi pengendalian. Penelitian ini bertujuan mengevaluasi pengaruh pemberian vaksin S. agalactiae hasil iradiasi yang dikombinasikan dengan nanopartikel kitosan terhadap morfometri folikel limfoid limpa mencit. Sebanyak 20 ekor mencit jantan galur DDY dibagi ke dalam lima kelompok perlakuan, yaitu Phosphate Buffered Saline (PBS), trehalosa, nanopartikel kitosan, vaksin iradiasi S. agalactiae, dan kombinasi vaksin dengan nanopartikel kitosan. Limpa dikoleksi empat hari pascaperlakuan, dibuat preparat histologis dengan pewarnaan hematoksilin-eosin, kemudian diamati jumlah dan ukuran folikel limfoid. Hasil penelitian menunjukkan bahwa kelompok kombinasi vaksin iradiasi S. agalactiae dan nanopartikel kitosan memiliki rerata jumlah folikel limfoid (16,00 ± 2,6) dan ukuran folikel limfoid (2438,2 ± 398,3 µm²) tertinggi serta berbeda nyata dibandingkan kelompok kontrol (p < 0,05). Temuan ini menunjukkan bahwa kombinasi nanopartikel kitosan dan vaksin iradiasi S. agalactiae mampu meningkatkan aktivitas jaringan limfoid limpa dan berpotensi meningkatkan imunogenisitas kandidat vaksin mastitis subklinis pada sapi perah.
       
      Subclinical mastitis is an important disease in dairy cattle that causes economic losses through reduced milk production and milk quality. One of the major causative agents is Streptococcus agalactiae. Although antibiotic therapy is commonly used for disease control, repeated administration may lead to antimicrobial resistance and drug residues. Therefore, vaccine development represents a promising alternative approach. This study aimed to evaluate the effect of an irradiated S. agalactiae vaccine combined with chitosan nanoparticles on the morphometry of splenic lymphoid follicles in mice. Twenty male DDY mice were randomly assigned to five treatment groups: Phosphate Buffered Saline (PBS), trehalose, chitosan nanoparticles, irradiated S. agalactiae vaccine, and a combination of the vaccine with chitosan nanoparticles. Spleens were collected four days after treatment, processed for hematoxylin-eosin staining, and examined for lymphoid follicle number and size. The combination group exhibited the highest mean lymphoid follicle number (16.00 ± 2.6) and follicle size (2438.2 ± 398.3 µm²), which were significantly greater than those of the control group (p < 0.05). These findings indicate that the combination of chitosan nanoparticles and irradiated S. agalactiae vaccine enhances splenic lymphoid activity and may improve the immunogenicity of a candidate vaccine against subclinical mastitis in dairy cattle
