dc.description.abstractPemeriksaan hemoglobin (Hb) merupakan parameter penting dalam menilai kondisi kesehatan hewan laboratorium. Penelitian ini bertujuan mengevaluasi stabilitas kadar Hb darah kelinci New Zealand White yang disimpan selama lima hari pada suhu 4 °C menggunakan antikoagulan EDTA pada berbagai tingkat konsentrasi. Variasi konsentrasi Hb dibuat secara in vitro menjadi tiga kelompok: rendah (75%), menengah (100%), dan tinggi (150%) melalui kombinasi whole blood, packed red blood cell (pRBC), dan NaCl fisiologis 0,9%. Kadar Hb diukur setiap hari menggunakan hematology analyzer Mindray BC-2800Vet dan dianalisis menggunakan uji two-way ANOVA. Hasil penelitian menunjukkan lama penyimpanan dan variasi konsentrasi berpengaruh signifikan (p < 0,05) terhadap kadar Hb. Hasil p-value interaksi antara variasi konsentrasi dan lama penyimpanan menunjukkan angka 0,419 (p > 0,05). Hal ini menunjukkan bahwa perubahan kadar Hb selama penyimpanan terjadi dengan pola yang serupa di seluruh kelompok konsentrasi, sehingga metode penyimpanan tetap aman digunakan pada berbagai kondisi klinis sampel selama lima hari.
dc.description.abstractHemoglobin (Hb) measurement is a critical parameter for assessing the health status of laboratory animals. This study aimed to evaluate the stability of Hb levels in New Zealand White rabbit blood stored for five days at 4 °C using EDTA anticoagulant across various concentration levels. Hb concentration variations were prepared in vitro into three groups: low (75%), medium (100%), and high (150%) through a combination of whole blood, packed red blood cells (pRBC), and 0,9% physiological saline. Hb levels were measured daily using a Mindray BC-2800Vet hematology analyzer and analyzed using a two-way ANOVA. The results showed that both storage duration and concentration variations significantly affected Hb levels (p < 0,05). The p-value for the interaction between concentration variation and storage duration was 0,419 (p > 0,05). Changes in Hb levels during storage followed a similar pattern across all concentration groups. This storage method remains safe for various clinical sample conditions for up to five days.
