      Abstract
      Imunohistokimia (IHK) merupakan teknik pewarnaan khusus untuk mendeteksi antigen spesifik dalam jaringan melalui reaksi antigen-antibodi. Keberhasilan visualisasi IHK sangat dipengaruhi oleh konsentrasi antibodi primer yang digunakan. Antibodi primer TNF-a brand Abcam® tipe ab6671 hanya memiliki rekomendasi pengenceran untuk jaringan manusia, sehingga optimasi diperlukan untuk aplikasinya pada jaringan tikus. Penelitian ini bertujuan menentukan pengenceran antibodi primer TNF-a yang paling optimal pada jaringan limpa tikus. Penelitian menggunakan preparat jaringan limpa tikus dari arsip blok parafin dengan tiga variasi pengenceran, yaitu 1:50, 1:100, dan 1:200, masing-masing dibuat tiga ulangan. Intensitas pewarnaan dianalisis menggunakan perangkat lunak Fiji yang menghasilkan nilai mean gray value, kemudian dikonversi menjadi nilai optical density (OD) dan dianalisis menggunakan uji One-Way ANOVA. Hasil menunjukkan nilai OD rata-rata sebesar 0,523 untuk pengenceran 1:50, 0,393 untuk 1:100, dan 0,221 untuk 1:200. Uji statistik tidak menunjukkan perbedaan signifikan antar kelompok (p=0,060). Pengenceran 1:100 menunjukkan keseimbangan terbaik secara kualitatif antara intensitas, spesifisitas, dan konsistensi pewarnaan, sehingga direkomendasikan sebagai pengenceran optimal untuk visualisasi TNF-a pada jaringan limpa tikus menggunakan antibodi Abcam® tipe ab6671.
       
      Immunohistochemistry (IHC) is a specialized staining technique used to detect specific antigens in tissue through antigen-antibody reactions. The Abcam® Anti-TNF-a antibody (catalog number ab6671) only includes dilution recommendations for human tissue, so optimization is required for its application in rat tissue. This study aims to determine the most optimal dilution of the TNF-a primary antibody in rat spleen tissue. The study used rat spleen tissue sections from a paraffin block archive with three dilution variations: 1:50, 1:100, and 1:200, each with three replicates. Staining intensity was analyzed using the Fiji software, which generated mean gray values, which were then converted into optical density (OD) values and analyzed using a one-way ANOVA. The results showed an average OD value of 0,523 for the 1:50 dilution, 0,393 for 1:100, and 0,221 for 1:200. Statistical analysis did not reveal significant differences between groups (p=0.060). The 1:100 dilution demonstrated the best qualitative balance between staining intensity, specificity, and consistency; therefore, it is recommended as the optimal dilution for visualizing TNF-a in rat spleen tissue using the Abcam® antibody type ab6671.
