      Perubahan Jumlah Makrofag Peritoneal terhadap Kandidat Vaksin Iradiasi Streptococcus agalactiae Kombinasi Nanopartikel Kitosan pada Mencit

      Abstract
      Mastitis subklinis yang disebabkan oleh Streptococcus agalactiae merupakan masalah penting pada sapi perah, sehingga diperlukan pengembangan vaksin yang mampu memicu aktivasi imun yang lebih efektif. Penelitian ini bertujuan mengevaluasi respons imun bawaan melalui dinamika makrofag peritoneal pada mencit setelah pemberian kandidat vaksin iradiasi S. agalactiae yang diformulasikan dengan nanopartikel kitosan sebagai adjuvan. Sebanyak 40 mencit galur DDY dibagi ke dalam sepuluh kelompok perlakuan dan diberikan injeksi intraperitoneal yang terdiri atas kontrol, nanopartikel kitosan, antigen iradiasi, serta kombinasi antigen–nanopartikel. Pengambilan cairan peritoneal dilakukan pada hari ke-1 dan ke-4 untuk menghitung jumlah makrofag sebagai indikator respons imun awal. Uji Two-Way ANOVA menunjukkan adanya interaksi signifikan antara jenis perlakuan dan waktu observasi (Sig. < 0,001). Pada hari ke-1, kombinasi antigen iradiasi dengan nanopartikel kitosan menghasilkan aktivasi makrofag peritoneal tertinggi, menunjukkan potensi kitosan dalam meningkatkan imunogenisitas antigen iradiasi. Pada hari ke-4, kelompok yang menerima antigen mengalami penurunan jumlah makrofag yang konsisten dengan fenomena Macrophage Disappearance Reaction, sedangkan kelompok kontrol tetap stabil. Temuan ini menegaskan bahwa formulasi nanopartikel kitosan mampu memperkuat respons imun bawaan dan berpotensi dikembangkan sebagai adjuvan inovatif dalam kandidat vaksin iradiasi S. agalactiae untuk pencegahan mastitis subklinis.
       
      Subclinical mastitis caused by Streptococcus agalactiae remains a major issue in dairy cows, highlighting the need for vaccines capable of inducing stronger immune activation. This study evaluated innate immune responses by examining the dynamics of peritoneal macrophages in mice following administration of an irradiated S. agalactiae vaccine candidate formulated with chitosan nanoparticles as an adjuvant. Forty DDY mice were allocated into ten treatment groups and received intraperitoneal injections consisting of a control, chitosan nanoparticles, irradiated antigen, and a combination of antigen–nanoparticles. Peritoneal fluid was collected on days 1 and 4 to quantify macrophage numbers as an indicator of early immune activation. Two-Way ANOVA revealed a significant interaction between treatment type and observation time (Sig. < 0.001). On day 1, the combination of irradiated antigen with chitosan nanoparticles triggered the highest peritoneal macrophage activation, demonstrating the ability of chitosan to enhance the immunogenicity of the irradiated antigen. On day 4, antigen-receiving groups showed a reduction in macrophage numbers consistent with the Macrophage Disappearance Reaction, while control groups remained stable. These findings confirm that chitosan nanoparticle formulation can potentiate innate immune activation and holds promise as an innovative adjuvant for S. agalactiae irradiated vaccine development against subclinical mastitis.
