| dc.description.abstract | Plasmodium sp. are protozoan parasites that cause malaria and infect both humans and non-human primates. This study aimed to optimize in vitro culture media for the growth of Plasmodium cynomolgi and Plasmodium inui derived from infected Macaca fascicularis blood samples. Cultures were maintained using RPMI-1640 supplemented with HEPES, D-glucose, and hypoxanthine, with two serum formulations evaluated: 20% Albumax and a combination of 10% fetal bovine serum (FBS) with 10% Albumax. Cultures were incubated at 36.5°C under controlled conditions (5% CO2, 5% O2, 90% N2), and parasite growth was monitored using Giemsa-stained thin blood smears, with parasitemia calculated every 48 hours. The results showed that the combination of 10% FBS and 10% Albumax was more effective in maintaining parasite survival compared to 20% Albumax alone, with the highest parasitemia observed in Plasmodium cynomolgi reaching 3.8% on day 6. These findings indicate that the culture conditions support short-term in vitro growth of Plasmodium and may serve as a basis for further development of malaria culture systems. | |
| dc.description.abstract | Plasmodium sp. merupakan protozoa penyebab malaria yang dapat menginfeksi manusia maupun primata non-manusia. Penelitian ini bertujuan untuk mengoptimasi media kultur in vitro untuk pertumbuhan Plasmodium cynomolgi dan Plasmodium inui yang berasal dari darah Macaca fascicularis terinfeksi. Kultur dilakukan menggunakan medium RPMI-1640 yang diperkaya dengan HEPES, D-glukosa, dan hipoksantin, dengan dua formulasi serum yaitu Albumax 20% dan kombinasi FBS 10% dengan Albumax 10%. Kultur diinkubasi pada suhu 36,5°C dengan komposisi gas 5% CO2, 5% O2, dan 90% N2, dan pertumbuhan parasit diamati melalui preparat ulas darah berwarna Giemsa serta dihitung berdasarkan parasitemia setiap 48 jam. Hasil menunjukkan bahwa kombinasi FBS 10% dan Albumax 10% lebih efektif dalam mempertahankan kelangsungan hidup parasit dibandingkan Albumax 20% saja, dengan parasitemia tertinggi ditemukan pada Plasmodium cynomolgi sebesar 3,8% pada hari ke-6. Hasil ini menunjukkan bahwa kondisi kultur mampu mendukung pertumbuhan jangka pendek Plasmodium secara in vitro dan dapat menjadi dasar pengembangan sistem kultur malaria selanjutnya. | |
