      Pembentukan Galur Padi Doubled Haploid melalui Kultur Antera untuk Mendukung Biofortifikasi dengan Zat Besi dan Zink

      Abstract
      Peningkatan gizi pada bahan pangan perlu dilakukan untuk mengurangi tingkat stunting dan pada anak-anak di Indonesia. Peningkatan kandungan gizi makanan pokok seperti padi melalui biofortifikasi menjadi cara terbaik untuk memenuhi kebutuhan gizi anak. Pembentukan galur padi yang memiliki kandungan zat besi dan zink menjadi salah satu solusi untuk penanganan stunting melalui sektor pertanian. Pemuliaan tanaman menggunakan kultur antera dapat dilakukan untuk mempercepat perolehan galur murni padi yang memiliki kandungan nutrisi tinggi. Tujuan umum pada penelitian ini adalah untuk mendapatkan galur-galur padi doubled haploid (DH) putatif yang memiliki kandungan zat besi dan zink melalui kultur antera. Kultur antera dilaksanakan di Bogor mulai bulan November 2024 sampai Agustus 2025. Materi genetik yang digunakan adalah empat genotipe F2 hasil persilangan padi yang memiliki kandungan Fe dan Zn tinggi yang digunakan sebagai sumber donor antera. Percobaan dilakukan dengan rancangan acak lengkap yang diulang sebanyak enam kali dengan genotipe donor antera sebagai faktor. Hasil penelitian menunjukkan bahwa sumber donor antera berpengaruh terhadap keberhasilan kultur antera. Genotipe berpengaruh terhadap keberhasilan antera dalam membentuk kalus, regenerasi tanaman hijau, dan regenerasi tanaman albino. Empat genotipe donor yang digunakan seluruhnya berhasil membentuk kalus dan beregenerasi menjadi tanaman. Genotipe donor D1 (Inpari IR Nutri Zinc × Biosalin 2 Agritan) dan D2 (Inpari IR Nutri Zinc × IIA5-9-2-1) menghasilkan tanaman albino saja sedangkan genotipe donor D3 (Inpari IR Nutri Zinc × Inpari 32 HDB) dan D4 (Inpari IR Nutri Zinc × IPB 9G) menghasilkan tanaman hijau dan tanaman albino. Tanaman hijau yang diperoleh selanjutnya diaklimatisasi. Terdapat 175 tanaman hijau sehat dan bebas kontaminasi yang diaklimatisasi, dan diperoleh 120 tanaman yang berhasil bertahan dalam proses aklimatisasi. Tanaman hijau hasil kultur antera memiliki tingkat ploidi yang bervariasi, sehingga diperlukan evaluasi untuk mendapatkan tanaman dengan ploidi doubled haploid. Tanaman hijau dari genotipe D3 (Inpari IR Nutri Zinc × Inpari 32 HDB) dan D4 (Inpari IR Nutri Zinc × IPB 9G) dievaluasi pada percobaan kedua untuk mengetahui tingkat ploidi dari masing-masing tanaman hijau. Percobaan dilaksanakan mulai bulan Juli 2025 hingga Januari 2026. Evaluasi dilakukan berdasarkan morfologi tanaman kemudian dikonfirmasi menggunakan pengamatan stomata dan flow cytometry. Setiap tanaman hijau diamati morfologinya untuk menapis tanaman haploid. Beberapa tanaman DH putatif kemudian dikonfirmasi menggunakan pengamatan stomata dan flow cytometry. Hasil pengamatan menunjukkan bahwa tanaman haploid dan tanaman DH memiliki morfologi yang berbeda. Ciri tanaman haploid yang paling mudah untuk dikenali adalah memiliki jumlah anakan yang banyak dan memiliki spikelet yang 100% steril, sedangkan tanaman DH memiliki penampilan yang menyerupai tetua diploid dengan spikelet fertil. Hasil pengamatan morfologi menunjukkan bahwa terdapat 26 tanaman DH putatif yang selanjutnya akan dikonfirmasi tingkat ploidinya menggunakan pengamatan stomata dan flow cytometry. Hasil pengamatan stomata menunjukkan bahwa tanaman haploid memiliki ukuran stomata lebih pendek dengan kerapatan stomata lebih tinggi dibandingkan dengan tetua diploid, sedangkan tanaman DH memiliki stomata yang panjang dan kerapatannya menyerupai tetua diploid. Konfirmasi menggunakan flow cytometry menunjukkan bahwa 26 galur tersebut memiliki ploidi doubled haploid.
       
      Improving the nutritional quality of food is necessary to reduce the prevalence of stunting among children in Indonesia. Enhancing the nutrient content of staple crops such as rice through biofortification is considered an effective approach to meet children's nutritional needs. The development of rice lines with high iron and zinc contents represents a potential solution for addressing stunting through the agricultural sector. Plant breeding using anther culture can be applied to accelerate the production of rice pure lines with high nutritional value. The general objective of this study was to obtain putative doubled haploid (DH) rice lines with high iron and zinc contents through anther culture. Anther culture was conducted in Bogor from November 2024 to August 2025. The genetic materials used consisted of four F2 genotypes derived from rice crosses with high Fe and Zn contents, which were utilized as anther donor sources. The experiment was arranged in a completely randomized design with six replications, using anther donor genotype as the experimental factor. The results showed that the anther donor source significantly affected the success of anther culture. Genotype influenced the ability of anthers to form callus, as well as the regeneration of green and albino plantlets. Among the four donor genotypes tested, all were capable of inducing callus formation and regenerating into plantlets. Donor genotypes D1 (Inpari IR Nutri Zinc × Biosalin 2 Agritan) and D2 (Inpari IR Nutri Zinc × IIA5-9-2-1) produced only albino plantlets, whereas donor genotypes D3 (Inpari IR Nutri Zinc × Inpari 32 HDB) and D4 (Inpari IR Nutri Zinc × IPB 9G) produced both green and albino plantlets. The regenerated green plants were subsequently acclimatized. A total of 175 healthy and contamination-free green plants were acclimatized, of which 120 plants successfully survived the acclimatization process. Green plants derived from anther culture exhibited varying ploidy levels; therefore, evaluation was required to obtain doubled haploid plants. Green plants from genotypes D3 (Inpari IR Nutri Zinc × Inpari 32 HDB) and D4 (Inpari IR Nutri Zinc × IPB 9G) were evaluated in the second experiment to determine the ploidy level of each plant. The experiment was conducted from July 2025 to January 2026. Evaluation was performed based on plant morphology and subsequently confirmed through stomatal observation and flow cytometry analysis. Each green plant was morphologically observed to screen for haploid plants. Several putative DH plants were then confirmed using stomatal observation and flow cytometry. The results showed that haploid and DH plants exhibited distinct morphological characteristics. The most easily recognizable characteristics of haploid plants were the production of numerous tillers resembling a bush and 100% sterile spikelets, whereas DH plants exhibited an appearance similar to the diploid parents with fertile spikelets. Morphological observations identified 26 putative DH plants, which were subsequently confirmed for their ploidy levels through stomatal observation and flow cytometry. Stomatal observations showed that haploid plants had shorter stomata with higher stomatal density compared with the diploid parents, whereas DH plants had stomatal size and density similar to those of the diploid parents. Confirmation using flow cytometry demonstrated that all 26 lines possessed doubled haploid ploidy.
