Burung kutilang (Pycnonotus aurigaster) merupakan burung liar yang berpotensi berperan dalam penyebaran penyakit unggas, termasuk coronavirus mirip Infectious Bronchitis Virus (IBV). Penelitian ini bertujuan mendeteksi
keberadaan coronavirus mirip IBV pada burung kutilang menggunakan metode reverse transcriptase polymerase chain reaction (RT-PCR). Sampel swab orofaring dan kloaka dikoleksi dari 10 ekor burung pada minggu ke-0, ke-2, dan ke-4. Amplifikasi dilakukan menggunakan primer XCE2F/XCE2R yang menargetkan gen S1 dengan ukuran fragmen 466 bp. Hasil menunjukkan seluruh
sampel minggu ke-0 dan ke-4 negatif, sedangkan pada minggu ke-2 terdeteksi pita DNA berukuran 466 bp pada pooling swab kloaka burung 1–5 dan pooling swab orofaring burung 6–10. Pengujian ulang terhadap sampel pooling positif dilakukan secara individual, namun tidak kembali menghasilkan pita DNA target. Temuan ini
mengindikasikan adanya coronavirus mirip IBV pada burung kutilang dan memberikan gambaran awal mengenai potensinya sebagai reservoir coronavirus
mirip IBV di lingkungan perunggasan. The Sooty-headed Bulbul (Pycnonotus aurigaster) is a wild bird that may play a role in the transmission of poultry diseases, including Infectious Bronchitis Virus
(IBV)-like coronavirus. This study aimed to detect the presence of an IBV-like coronavirus in Sooty-headed Bulbuls using transcriptase polymerase chain reaction (RT-PCR). Oropharyngeal and cloacal swab samples were collected from ten birds at weeks 0, 2, and 4. Amplification was performed using the XCE2F/XCE2R primer pair targeting the S1 gene, with an expected amplicon size of 466 bp. The results showed that all samples collected at weeks 0 and 4 were negative, whereas at week 2, a 466 bp DNA band was detected in pooled cloacal swab samples from birds 1–5 and pooled oropharyngeal swab samples from birds 6–10. Re-testing of the positive pooled samples was subsequently performed on individual samples; however, no target DNA band was detected. These findings indicate the presence of an IBV-like coronavirus in Sooty-headed Bulbuls and
provide preliminary evidence of their potential role as a reservoir of IBV-like coronavirus in poultry environments.
