      Parasitemia Profile and Blood Smear Analysis of Plasmodium sp. in Macaca fascicularis Infected with Malaria.

      Abstract
      Malaria is a parasitic disease caused by Plasmodium spp. and transmitted by female Anopheles mosquitoes. Several Plasmodium species infect non-human primates, including Macaca fascicularis, which serve as natural reservoirs and contribute to the epidemiology of zoonotic malaria. This study aimed to analyze the parasitemia profile and blood smear characteristics of Plasmodium infection in M. fascicularis and to confirm the infecting species using molecular techniques. A total of 71 blood samples were examined by Giemsa-stained thin blood smear microscopy to evaluate parasite morphology and parasitemia, followed by conventional PCR targeting the SSU rRNA gene and Sanger sequencing. Microscopic examination predominantly identified early trophozoite stages. Parasitemia analysis showed that 11 samples had low parasitemia (mean 1.6%), whereas five samples exhibited high parasitemia (mean 6.85%). PCR detected Plasmodium DNA in 26 samples, demonstrating greater sensitivity than microscopy. Sequencing of nine PCR-positive samples identified Plasmodium cf. inui with 94–97% sequence identity. These findings indicate that most infections in M. fascicularis are low-density and likely chronic, supporting their role as natural reservoir hosts while highlighting the superior sensitivity of molecular methods for detecting submicroscopic infections. This study provides baseline information for simian malaria surveillance, improved diagnostic approaches, and future research on zoonotic malaria transmission and antimalarial drug development.
       
      Malaria merupakan penyakit parasit yang disebabkan oleh Plasmodium spp. dan ditularkan melalui gigitan nyamuk Anopheles betina. Beberapa spesies Plasmodium menginfeksi primata non-manusia, termasuk Macaca fascicularis, yang berperan sebagai reservoir alami dan berpotensi mendukung penularan malaria zoonotik. Penelitian ini bertujuan menganalisis profil parasitemia dan karakteristik ulas darah infeksi Plasmodium pada M. fascicularis, serta mengkonfirmasi spesies penyebab infeksi menggunakan metode molekuler. Sebanyak 71 sampel darah diperiksa menggunakan mikroskopi ulas darah tipis yang diwarnai Giemsa untuk mengamati morfologi parasit dan menghitung tingkat parasitemia, kemudian dilanjutkan dengan PCR konvensional yang menargetkan gen SSU rRNA dan analisis sekuensing Sanger. Hasil pemeriksaan mikroskopis menunjukkan dominasi stadium trofozoit awal. Analisis parasitemia menunjukkan bahwa 11 sampel memiliki parasitemia rendah (rata-rata 1,6%) sedangkan lima sampel memiliki parasitemia tinggi (rata-rata 6,85%). Pemeriksaan PCR mendeteksi DNA Plasmodium pada 26 sampel, menunjukkan sensitivitas yang lebih tinggi dibandingkan mikroskopi. Analisis sekuensing terhadap sembilan sampel positif PCR mengidentifikasi Plasmodium cf. inui dengan tingkat kemiripan sekuens sebesar 94–97%. Hasil penelitian ini menunjukkan bahwa sebagian besar infeksi pada M. fascicularis bersifat kronis dengan parasitemia rendah, sehingga mendukung perannya sebagai reservoir alami malaria simian. Penelitian ini memberikan informasi dasar yang bermanfaat bagi surveilans malaria simian, peningkatan akurasi diagnosis, serta pengembangan penelitian mengenai penularan malaria zoonotik dan pengembangan obat antimalaria.
