Fusarium oxysporum merupakan cendawan fitopatogen yang menginfeksi tanaman pertanian melalui tanah. Infeksi Fusarium pada tanaman kedelai mencapai 60% dan mengakibatkan kerugian ekonomi yang besar bagi petani kedelai setiap tahun di seluruh dunia. Dinding sel cendawan tersebut mengandung 88–90% ß-glukan sehingga sulit untuk dihambat. Penggunaan fungisida sintetik dalam mengatasi infeksi tersebut dilaporkan berdampak buruk bagi lingkungan dan meningkatkan resisten patogen. Maka dari itu, untuk mengatasi hal tersebut dihadirkan Streptomyces sebagai mikrob rizosfer menjadi agens pengendali hayati bersifat ramah lingkungan yang memiliki kemampuan menghasilkan enzim ß-1,3-glukanase. Enzim ß-1,3-glukanase mampu menghidrolisis ikatan ß-1,3-glikosidik pada glukan yang menyusun dinding sel cendawan. Penggunaan Streptomyces yang sudah diketahui potensinya pada penelitian sebelumnya sebagai penghasil enzim glukanase, menginisiasi penelitian ini untuk mengetahui aktivitas Streptomyces pada tanah perakaran (rizosfer) tanaman kedelai dalam menghambat aktivitas cendawan fitopatogen F. oxysporum melalui aplikasi in planta. Penelitian ini bertujuan untuk mengevaluasi kemampuan Streptomyces penghasil enzim glukanase dalam mengendalikan penyakit busuk akar yang disebabkan F. oxysporum dan menganalisis profil mikrobiom rizosfer kedelai.
Penelitian ini dilakukan melalui beberapa prosedur kerja meliputi peremajaan ulang koleksi Streptomyces spp. yang sudah berpotensi sebagai penghasil enzim glukanase. Uji verifikasi terhadap Streptomyces spp. terkait aktivitas glukanolitik dan antifungalnya. Evaluasi virulensi dari F. oxysporum terhadap benih kedelai. Pengamatan insidensi dan reduksi penyakit pada tanaman kedelai dilakukan secara in planta di rumah kaca, sedangkan untuk analisis metagenom amplikon 16S rRNA untuk mengetahui profil mikrobiom rizosfer dipilih sampel tanah rizosfer dari tanaman sakit dan sehat dari perlakuan yang sudah diberi formula terbaik. Data terkait insidensi penyakit, reduksi penyakit ditabulasi pada Microsoft Excel 2021, dilanjutkan uji Duncan dengan tingkat kepercayaan 95% untuk uji di laboratorium. Analisis secara statistik dengan ANOVA two-way dan uji Tukey pada taraf nyata 5% untuk data dari pengamatan di rumah kaca dan visualisasi data menggunakan RStudio. Analisis metagenom menggunakan Dorado, NanoPlot, dan RStudio (R versi 4.3).
Streptomyces spp. dengan kode ARJ 32, ARJ 38, ARJ 42, dan ARJ 44 memiliki aktivitas glukanolitik dan antifungi dengan aktivitas tertinggi diamati pada isolat Streptomyces ARJ 32 dan ARJ 44. Media alternatif terpilih yang digunakan perbanyakan biomassa sel Streptomyces ARJ 32 dan ARJ 44 sebagai bahan yang dicampur pada formula padat gambut adalah ekstrak ubi jalar dengan biomassa kering pada kisaran 4,11 - 4,48 mg/ml. Empat kemasan formula dengan kode F32G, F32NG, F44G, dan F44NG mampu mempertahankan vibilitas sel Streptomyces dengan konsentrasi 4,0 x 106 - 8,0 x 107 CFU g-1 yang disimpan selama 38 minggu. Aplikasi in planta memperoleh dua formula terbaik yaitu F32NG dan F44G yang efektif dan signifikan pada tanah non steril dalam menurunkan insidensi penyakit dengan tingkat infeksi sebesar 54% menjadi 4% yang diberi formula F32NG dan 12% yang diberi formula F44G, sehingga mampu mereduksi penyakit pada tanaman kedelai yang terinfeksi F. oxysporum 92% pada formula F32NG dan 77% pada formula F44G.
F32NG yang mengandung kultur dari S. tendae ARJ 32 menjadi formula terbaik yang mampu memperbaiki pertumbuhan rambut akar kedelai tanpa menunjukkan tilosis pada pembuluh xilem akar melalui pengamatan Scanning Electron Microscope (SEM). Pemberian formula F32NG sebagai agens biokontrol F. oxysporum mampu berinteraksi secara sinergis dengan mikrobiom rizosfer asli dan membentuk struktur mikrobiom. Modulasi ini meningkatkan kelimpahan bakteri pemacu tumbuh tanaman (PGPR), sehingga mendukung ketahanan dan kesehatan tanaman secara keseluruhan. Temuan tersebut menegaskan bahwa formula S. tendae ARJ 32 berpotensi sebagai agens biokontrol cendawan fitopatogen F. oxysporum yang efektif, berkelanjutan, sinergis, dan stabil di tanah alami melalui aktivitas antifungi dan memengaruhi struktur profil komunitas mikrobiom rizosfer.

Fusarium oxysporum is a phytopathogenic fungus that infects agricultural crops through the soil. Fusarium infections in soybean crops reach 60% and result in significant economic losses for soybean farmers worldwide each year. The fungal cell wall contains 88–90% ß-glucan, making it difficult to inhibit. The use of synthetic fungicides to control this infection has been reported to negatively impact the environment and increase pathogen resistance. Therefore, to overcome this issue, Streptomyces, a rhizosphere microbe, is introduced as an environmentally friendly biocontrol agent capable of producing the enzyme ß-1.3-glucan. The ß-1.3-glucanase enzyme is capable of hydrolyzing the ß-1.3-glycosidic bonds in glucans in these cendawan. The use of Streptomyces, whose potential as a glucanase producer has been recognized in previous research, prompted this study to determine the activity of Streptomyces in the rhizosphere of soybean plants in inhibiting the phytopathogenic fungus F. oxysporum through in-plant application. This study aimed to evaluate the ability of glucanase-producing Streptomyces to control root rot caused by F. oxysporum and analyze the soybean rhizosphere microbiome profile.
This research was conducted through several work procedures, including rejuvenation of a collection of Streptomyces spp. that already have potential as glucanase enzyme producers. Verification tests on Streptomyces spp. for their glucanolytic and antifungal activities were conducted on Streptomyces spp., and evaluation of the virulence of F. oxysporum on soybean seedlings. Observations of disease incidence and reduction in soybean plants were conducted in planta in a greenhouse. 16S rRNA amplicon metagenome analysis for knowing rhizosphere microbiome profiles included rhizosphere soil samples from diseased and healthy plants treated with the best formula. Data related to disease incidence and disease reduction tabulated in Microsoft Excel 2021. Duncan's test with a 95% confidence level for laboratory testing. Statistical analysis was performed using two-way ANOVA and Tukey's test at a 5% significance level for greenhouse observations and the visualization of data used RStudio. Dorado, NanoPlot, and RStudio (R version 4.3) were used for metagenome analysis.
Streptomyces spp. with codes ARJ 32, ARJ 38, ARJ 42, and ARJ 44 had glucanolytic and antifungal activities with the highest activity observed in Streptomyces isolates ARJ 32 and ARJ 44. The selected alternative medium used to produce high yields of Streptomyces ARJ 32 and ARJ 44 cell biomass which was then mixed into a peat-based solid formulation was sweet potato extract, yielding dry biomass in the range of 4.11–4.48 mg/mL. Four formula packages with codes F32G, F32NG, F44G, and F44NG were able to maintain the viability of Streptomyces cells with a concentration of 4.0 x 106- 8.0 x 107 CFU g-1 which was stored for 38 weeks. In planta application obtained two best formulas: F32NG and F44G. Both were effective and significant in non-sterile soil at reducing disease incidence. The F32NG formula reduced disease incidence from 54% to 4%, while the F44G formula reduced it to 12%, so that it was able to reduce disease in soybean plants infected with F. oxysporum 92% in the F32NG formula and 77% in the F44G formula.
F32NG containing culture of S. tendae ARJ 32 was the best formula that was able to improve the growth of soybean root hairs without showing tyloses in the root xylem vessels through Scanning Electron Microscope (SEM) observation. The applacation of S. tendae F32NG as a F. oxysporum biocontrol agent synergistically interacts with the indigenous rhizosphere microbiome and shapes the rhizosphere microbiome structure. This modulation promotes the abundance of plant growth-promoting rhizobacteria (PGPR), thereby supporting overall plant resistance and health. The findings confirm that the S. tendae ARJ 32 has the potential to be an effective, sustainable, synergistic, and stable biocontrol agent for the phytopathogenic fungus F. oxysporum in natural soil through antifungal activity and influencing the structure of the rhizosphere microbiome community profile.
