dc.description.abstractFeline coronavirus (FCoV) bersifat endemik pada kucing di Indonesia dan dapat memicu manifestasi penyakit yang sulit diprediksi. Penelitian ini bertujuan melakukan karakterisasi molekuler dan analisis filogenetik FCoV isolat Tangerang, Indonesia berdasarkan gen Nucleocapsid (N). Sebelas sampel arsip yang terkonfirmasi seluruhnya positif FCoV berdasarkan gen 3' untranslated region (UTR) dan sepuluh yang positif FCoV gen Spike (S) dianalisis menggunakan RT-PCR dengan target gen N, kemudian dilakukan sekuensing dan analisis filogenetik untuk memetakan hubungan kekerabatannya dengan referensi global. Sebanyak tujuh dari sebelas sampel tersebut menunjukkan hasil positif gen N FCoV dan berdasarkan analisis sekuens menunjukkan homologi nukleotida yang tinggi dengan FCoV genotipe 1, serta memiliki kedekatan kekerabatan dengan isolat referensi dari UK, USA, China, dan isolat Indonesia lain yang sudah dilaporkan sebelumnya. Klasterisasi tujuh sampel menunjukkan evolusi genetik spesifik pada isolat lokal. Sifat konservatif gen N pada pohon filogenetik berpotensi mendukung pengembangan vaksin serta pentingnya karakterisasi molekuler FCoV di Indonesia.
dc.description.abstractFeline coronavirus (FCoV) is endemic in Indonesian cats and can cause unpredictable clinical manifestations. This study aimed to perform molecular characterization and phylogenetic analysis of FCoV isolates from Tangerang, Indonesia, based on the Nucleocapsid (N) gene. Eleven archive samples that tested positive for FCoV based on the 3' untranslated region (UTR) gene and ten that tested positive for FCoV based on the Spike (S) gene were analyzed using RT-PCR targeting the N gene, followed by sequencing and phylogenetic analysis to map their phylogenetic relationships with global references. Seven out of the eleven samples tested positive for the FCoV N gene, and sequence analysis revealed high nucleotide homology with FCoV genotype 1, as well as close phylogenetic relationships with reference isolates from the UK, the USA, China, and other previously reported Indonesian isolates. The clustering of the seven samples reveals specific genetic evolution in local isolates. The conservative nature of the N gene on the phylogenetic tree potentially supports vaccine development and underscores the importance of molecular characterization of FCoV in Indonesia.
