      Abstract

Sengon (Falcataria falcata) is a fast-growing plant with high ecological and economic value. However, its productivity and availability have begun to decline due to pest attacks, especially boktor larvae. This study aims to determine the effect of elevation differences on the intensity of boktor pest attacks on sengon plants in Central Java, analyzing the association between SNP markers and sengon DNA accessions that are resistant and susceptible to boktor pests. Samples were collected by the research team at BKPH Kebon, KPH Mantingan, Rembang Regency and the Community Forest of Noborejo Village, Salatiga City. A total of 100 samples were tested, with details of 25 pairs of susceptible and resistant plants in each location. Sample testing was carried out using primers AAI_1, AAI_2, TI_1, TI_2 with the High Resolution Melting (HRM) method. The results showed that the attack intensity at both locations was relatively similar, namely 38% in Rembang and 39.2% in Salatiga. The Mann-Whitney test results showed that elevation did not significantly affect the attack intensity. The chi-square test showed that all primers had significant associations with sengon resistance to boktor pests (?^2 count > ?^2 table). Principal Component Analysis (PCA) analysis showed overlap between the resistant and susceptible groups, although some accessions showed clear genetic differentiation. The AAI_1 marker showed consistent associations at both elevations that can be used as a candidate molecular marker for sengon selection for boktor pest resistance. Accessions RR22 and RR25 with the CC genotype are recommended as candidate trees.
