      Aplikasi Bakteriofage sebagai Upaya Pengobatan Penyakit Motile Aeromonas Septicemia (MAS) pada Ikan Nila Oreochromis niloticus

      Abstract
      Intensifikasi budidaya seringkali dihadapkan pada serangan penyakit yang bergantung pada antibiotik, sehingga diperlukan alternatif biologis yang spesifik dan aman yaitu dengan bakteriofage. Penelitian ini bertujuan untuk mengaplikasikan bakteriofage sebagai agen biologis yang efektif dan aman dalam pengobatan penyakit Motile Aeromonas Septicemia (MAS) pada ikan nila. Penelitian ini diawali dengan persiapan bakteri uji, uji plak bakteriofage, uji penghambatan bakteri, dan pemeliharaan ikan. Parameter yang diamati meliputi karakteristik serta kepadatan bakteriofage, nilai Multiplicity of Infection (MOI), gejala klinis, dan hematologi ikan. Rancangan penelitian terdiri dari kontrol negatif (K), kontrol positif (K+), perlakuan antibiotik (AN), dan perlakuan bakteriofage (BK). Hasil penelitian menunjukkan bahwa bakteriofage berbentuk irregular, bulat dan memanjang yang berdiameter sekitar 2,51±0,12 mm. Kepadatan bakteriofage yang diperoleh 3,17×108 PFU mL?¹ dengan nilai Multiplicity of Infection (MOI) sebesar 2,34 bakteriofage sel-1. Bakteriofage mampu menekan pertumbuhan A. hydrophila ditandai dengan gejala klinis yang mulai membaik, diikuti dengan peningkatan hematologi pada ikan.
       
      Intensifying cultivation often faces disease outbreaks that depend on antibiotics, so a specific and safe biological alternative is needed, namely bacteriophages. This study aimed to apply bacteriophages as an effective and safe biological agent in treating Motile Aeromonas Septicemia (MAS) in tilapia. The research began with the preparation of test bacteria, bacteriophage plaque tests, bacterial inhibition tests, and fish maintenance. Observed parameters included the characteristics and density of bacteriophages, Multiplicity of Infection (MOI) values, clinical symptoms, and fish hematology. The study design included a negative control (K), a positive control (K+), an antibiotic treatment (AN), and a bacteriophage treatment (BK). The results showed that the bacteriophages are irregular, round, and elongated in shape with a diameter of about 2.51±0.12 mm. The density of bacteriophages obtained was 3.17×108 PFU mL?¹ with a Multiplicity of Infection (MOI) value of 2.34 phages per cell. The bacteriophages were able to suppress the growth of A. hydrophila, indicated by improving clinical symptoms, followed by an increase in the fish's hematology.
