      Variasi Morfologi dan Genetik Macrobrachium pilimanus Species Group di Jawa dan Sumatra

      Abstract
      Macrobrachium pilimanus species group menunjukkan keragaman morfologi yang tinggi, sehingga identifikasi spesies dan penentuan batas spesies dalam kelompok ini masih menjadi tantangan dalam kajian taksonomi. Distribusinya yang luas di Indonesia juga berpotensi mendorong diferensiasi antar populasi, sehingga semakin menyulitkan delimitasi spesies. Oleh karena itu, diperlukan pendekatan integratif yang menggabungkan data morfologi dan genetik untuk mengevaluasi variasi serta status taksonomi kelompok ini. Penelitian ini bertujuan mengkaji variasi morfologi dan genetik pada Macrobrachium pilimanus species group. Dalam penelitian ini dilakukan analisis morfometri geometrik yang berfokus pada karakter karapas dan pereiopod kedua. Selanjutnya, analisis genetik dilakukan menggunakan penanda mitokondria (COI dan 16S) serta penanda inti (28S). Variasi morfometri dianalisis menggunakan PCA, relative warp, dan uji korelasi Procrustes, sementara analisis genetik meliputi rekonstruksi filogenetik, perhitungan jarak genetik, serta identifikasi karakter sekuens diagnostik. Analisis morfometri menunjukkan adanya tumpang tindih sebagian antar populasi. Hasil molekuler mengidentifikasi beberapa garis keturunan genetik utama yang menunjukkan adanya keterkaitan antarwilayah sekaligus divergensi di dalam wilayah. Selain itu, uji korelasi menunjukkan hubungan yang lemah antara jarak morfologi dan jarak genetik. Secara keseluruhan, temuan ini menunjukkan pola variasi yang kompleks serta adanya potensi spesiasi pada Macrobrachium pilimanus species group.
       
      Macrobrachium pilimanus species group exhibits high morphological diversity, making species identification and species delimitation within this group a persistent challenge in taxonomic studies. Its wide distribution across Indonesia may also promote population differentiation, further complicating species delimitation. Therefore, an integrative approach combining morphological and genetic data is required to evaluate variation and clarify the taxonomic status of this group. This study aimed to investigate morphological and genetic variation within the Macrobrachium pilimanus species group. In this study, geometric morphometric analyses were conducted focusing on carapace characters and the second pereiopod. Subsequently, genetic analyses were performed using mitochondrial markers (COI and 16S) as well as a nuclear marker (28S). Morphometric variation was analyzed using PCA, relative warps, and procrustes correlation tests, while genetic analyses included phylogenetic reconstruction, genetic distance calculations, and identification of diagnostic sequence characters. Morphometric analyses revealed partial overlap among populations. Molecular results identified several major genetic lineages, indicating both inter-regional connectivity and intra-regional divergence. Furthermore, the correlation test revealed a weak relationship between morphological and genetic distances. Overall, these findings suggest a complex pattern of variation and the potential for speciation within the Macrobrachium pilimanus species group.
