dc.description.abstractRamin (Gonystylus bancanus) merupakan spesies pohon endemik hutan rawa gambut Asia Tenggara yang terancam punah, sehingga informasi genetiknya esensial untuk mendukung upaya konservasi. Penelitian ini bertujuan merakit dan menganalisis genom kloroplas G. bancanus serta menentukan posisi filogenetiknya di dalam famili Thymelaeaceae. Pengurutan dilakukan dengan long-read sequencing MinION Oxford Nanopore Technologies (ONT). Sebanyak 98.961 reads kloroplas berhasil diekstraksi dari 36,07 Gb data mentah. Hasil anotasi menunjukkan struktur sirkuler kuadripartit sepanjang 176.568 bp yang terdiri atas wilayah large single copy (89.005 bp), small single copy (3.372 bp), dan sepasang inverted repeat (±42.096 bp) dengan kandungan GC sebesar 35,92%. Fitur utama genom ini adalah terjadinya ekspansi pada wilayah inverted repeat. Analisis filogenetik memisahkan G. bancanus secara tegas dari subfamili Thymelaeoideae. Informasi molekuler genom kloroplas yang dihasilkan bermanfaat sebagai landasan upaya perlindungan pemanfaatan kayu ramin.
dc.description.abstractRamin (Gonystylus bancanus) is an endangered tree species endemic to Southeast Asia's peat swamp forests, making its genetic information essential to supporting conservation efforts. This study aims to assemble and analyze the chloroplast genome of G. bancanus and determine its phylogenetic position within the Thymelaeaceae family. Sequencing was performed using long-read sequencing with the MinION from Oxford Nanopore Technologies (ONT). A total of 98,961 chloroplast reads were successfully extracted from 36.07 Gb of raw data. Annotation results revealed a quadripartite circular structure spanning 176,568 bp, comprising a large single copy region (89,005 bp), a small single copy region (3,372 bp), and a pair of inverted repeats (±42,096 bp), with a GC content of 35.92%. A key feature of this genome is the expansion of the inverted repeat region. Phylogenetic analysis clearly separates G. bancanus from the subfamily Thymelaeoideae. The molecular information derived from the chloroplast genome serves as a foundation for efforts to protect the sustainable use of ramin wood.
