      Sintesis Hidrogel Berbasis Lignin Organosolv dan Potensinya sebagai Pembalut Luka

      Abstract
      Daun tebu merupakan biomassa lignoselulosa hasil samping perkebunan tebu yang belum banyak dimanfaatkan. Pemanfaatan daun tebu sebagai sumber lignin menawarkan strategi yang menjanjikan untuk memperluas aplikasinya dalam pengembangan bahan fungsional berbasis lignin. Lignin memiliki karakteristik kimia yang sesuai untuk aplikasi biomedis, seperti formulasi hidrogel pembalut luka. Tantangan dalam proses konversi dan karakteristik lignin dari daun tebu adalah kadar abu yang tinggi, sehingga diperlukan modifikasi proses antara lain dengan pra-perlakuan pencucian. Selain itu, lignosulfonat (lignin termodifikasi) secara luas digunakan sebagai aditif dalam hidrogel pembalut luka; namun, kekhawatiran tentang isu keberlanjutan dan potensi toksisitas bagi manusia membatasi penggunaannya. Penelitian ini bertujuan untuk mensintesis hidrogel berbasis lignin organosolv dari daun tebu sebagai pendekatan inovatif dalam perawatan luka. Lignin diekstraksi dengan metode organosolv etanol, dan hidrogel disiapkan dengan metode pengikat silang fisik melalui siklus beku-cair sebagai alternatif yang lebih ramah lingkungan. Pra-perlakuan pencucian dengan air dan asam asetat dilakukan sebelum proses ekstraksi untuk meminimalkan dampak pengolahan biomassa. Hasil menunjukkan bahwa pencucian daun tebu menurunkan kadar abu tanpa menyebabkan perubahan substansial pada struktur dan gugus fungsi daun tebu. Penurunan kadar abu tertinggi (49,93%) diperoleh pada perlakuan asam asetat 1M suhu 95 °C (DT-As95). Daun tebu tanpa pra-perlakuan (DT RAW) dan DT-As95 dipilih untuk proses ekstraksi lignin. Pra-perlakuan pencucian daun tebu efektif meningkatkan kemurnian lignin, dan lignin As (DT-As95) mencapai kemurnian 86,19 %, lebih tinggi dibandingkan dengan lignin R (DT-RAW) sebesar 73,53 %. Lignin hasil ekstraksi menunjukkan kemurnian yang relatif tinggi dengan aktivitas antioksidan dan antibakteri yang baik, sehingga berpotensi untuk dikembangkan dalam formulasi hidrogel pembalut luka. Penambahan lignin organosolv daun tebu ke dalam matriks PVA-kitosan meningkatkan kapasitas pembengkakan, kekuatan mekanis, dan aktivitas antioksidan hidrogel, serta memberikan kemampuan penyerapan sinar UV dan aktivitas antibakteri yang baik. Berdasarkan penelitian ini, penambahan lignin As menunjukkan sifat hidrogel yang lebih baik dibandingkan dengan lignin R, menegaskan potensi lignin organosolv daun tebu sebagai aditif dalam sistem hidrogel untuk aplikasi pembalut luka.
       
      Sugarcane leaves are lignocellulosic biomass derived as a by-product of sugarcane cultivation that remain underutilized. Their use as a lignin source offers a promising strategy to expand lignin applications in the development of functional materials. Lignin possesses chemical characteristics suitable for biomedical applications, such as hydrogel wound dressings. However, the high ash content of sugarcane leaf lignin presents a major challenge, necessitating process modifications including leaching pretreatments. In addition, lignosulfonates (modified lignin) are widely utilized as an additive in hydrogel wound dressings; however, sustainability concerns and possible human toxicity limit their use. This study aims to synthesize organosolv lignin-based hydrogels from sugarcane leaves as an innovative approach to wound care. Lignin was extracted using the organosolv method with ethanol as a solvent, and hydrogels were prepared via physical crosslinking through freeze-thaw cycles as a more environmentally friendly alternative. Leaching pretreatments with water and acetic acid were conducted prior to extraction to minimize biomass processing impacts. The results indicated that leaching effectively reduced the ash content while maintaining the structural and functional integrity of the sugarcane leaves. The highest ash reduction (49.93%) was achieved with 1 M acetic acid treatment at 95°C (DT-As95). Sugarcane leaves without pretreatment (DT-RAW) and DT-As95 were selected for lignin extraction. Leaching pretreatment significantly improved lignin purity, with lignin As (DT-As95) reaching 86.19%, higher than lignin R (DT-RAW) at 73.53%. The extracted lignin demonstrated high purity and showed notable antioxidant and antibacterial activities, suggesting its potential use in hydrogel formulations for wound dressings. The incorporation of sugarcane leaf organosolv lignin into the PVA-chitosan matrix improved the hydrogel's swelling capacity, mechanical strength, and antioxidant activity. Additionally, it provided UV absorption and antibacterial properties. Based on these findings, lignin As demonstrated superior hydrogel performance compared to lignin R, confirming the potential of sugarcane leaf organosolv lignin as an additive in hydrogel systems for wound dressing applications.
