      Teknik Sterilisasi Eksplan Biji Aren (Arenga pinnata) untuk Penyediaan Bahan Baku Bibit secara in Vitro

      Abstract
      Aren (Arenga pinnata (Wurmb) Merr.) memiliki fungsi ekologis dan ekonomis yang tinggi. Kultur jaringan dapat menjadi salah satu cara budidaya secara in vitro yang mengurangi permasalahan pada metode konvensional. Penelitian bertujuan mengidentifikasi teknik sterilisasi yang dapat digunakan pada kultur jaringan aren, mengidentifikasi teknik sterilisasi yang paling efektif untuk mendapatkan bahan baku bibit aren yang steril, dan menganalisis pengaruh masing-masing teknik sterilisasi terhadap tingkat kontaminasi aren. Penelitian dirancang dengan menggunakan rancangan acak lengkap satu faktor dengan jenis eksplan biji. Faktor yang dipakai terdiri dari tiga jenis perlakuan eksplan yaitu eksplan tidak dibakar dan tidak diberikan O3 (M0), eksplan dibakar dan tidak diberikan O3 (M1), serta eksplan tidak dibakar dan diberikan O3 (M2). Hasil penelitian menunjukkan ketiga perlakuan dapat digunakan pada kultur jaringan tanaman aren. Perlakuan M1 merupakan teknik sterilisasi yang paling efektif untuk mendapatkan bahan baku bibit aren yang steril. Perlakuan M1 juga memiliki tingkat kontaminasi yang paling rendah.
       
      Sugar palm (Arenga pinnata (Wurmb) Merr.) has high ecological and economic functions. Tissue culture can be one of the in vitro cultivation methods that reduce problems in conventional methods. The study aims to identify sterilization techniques that can be used in sugar palm tissue culture, identify the most effective sterilization technique to obtain sterile sugar palm seed raw materials, and analyze the effect of each sterilization technique on the level of sugar palm contamination. The study was designed using a completely randomized design with one factor with the type of seed explant. The factors used consisted of three types of explant treatments: explants not burned and not given O3 (M0), explants burned and not given O3 (M1), and explants not burned and given O3 (M2). The results showed that all three treatments can be used in sugar palm tissue culture. Treatment M1 is the most effective sterilization technique to obtain sterile sugar palm seed raw materials. Treatment M1 also has the lowest level of contamination.
