Evaluasi Kualitas, Kinematik, dan Freezeability Semen Sapi Belgian Blue dalam Berbagai Pengencer

Penelitian ini bertujuan untuk mengkarakterisasi kualitas semen segar, menganalisis profil biokimia plasma semen dan membandingkan pengencer buatan sendiri (homemade) dan pengencer komersial untuk pembekuan semen sapi bergian blue. Semen dikoleksi dari 4 ekor sapi Belgian blue milik BIB Lembang. Semen dievaluasi dan masing-masing dibagi 4 pengencer Tris Kuning Telur dan Pengencer komersial (AndroMed®, BoviFree®, dan Steridyl®). Semen dikemas ke dalam straw dan diequilibrasi serta dibekukan. Hasil analisis menunjukkan viabilitas, integritas akrosom, dan abnormalitas sperma pada semen segar sapi belgian blue tidak berbeda (p>0,05). Biokimia plasma semen sapi bergian blue mengandung fruktosa (616,09-961,61 mg/dl), MDA (2,3-33,28 nmol/ml), TAC (8-44%), GOT (261-632 U/l), GPT (9-20 U/l), TP (6,4-13,9 g/dl), albumin (1,6-3,9 g/dl), kolesterol (18,5-33,4 mg/dl), trigliserida (18,7-52,4 mg/dl). Motilitas dan viabilitas sperma pengencer tris kuning telur gliserol lebih baik daripada tris kuning telur DMF (p<0,05). Kinematik sperma menunjukkan tidak ada perbedaan antara pengencer homemade dan pengencer komersial. TKTG menunjukkan kinematik dan motilitas sperma yang lebih tinggi dibandingkan dengan pengencer lainnya. Nilai Recovery rate (RR) tidak berbeda nyata antar pengencer. Longevitas sperma dalam pengencer BoviFree® paling baik dapat dipertahankan motilitas sperma selama 3 jam penyimpanan (24,65±5,53) dan 4 jam penyimpanan (19,69±4,50). This study aimed to characterise the quality of fresh semen, analyse the biochemical profile of seminal plasma, and compare homemade and commercial diluents for freezing Belgian Blue cattle semen. Semen was collected from four Belgian Blue cattle owned by BIB Lembang. The semen was evaluated and divided into four diluents (Tris Egg Yolk Diluent and commercial diluents (AndroMed®, BoviFree®, and Steridyl®) each. Semen was packed into straws, equilibrated, and frozen. The results showed no differences in viability, acrosome integrity, and sperm abnormalities in fresh Belgian Blue cattle semen (p>0.05). Biochemistry of Belgian Blue cattle semen plasma contains fructose (616.09-961.61 mg/dl), MDA (2.3-33.28 nmol/ml), TAC (8-44%), GOT (261-632 U/l), GPT (9-20 U/l), TP (6.4-13.9 g/dl), albumin (1.6-3.9 g/dl), cholesterol (18.5-33.4 mg/dl), triglycerides (18.7-52.4 mg/dl). Sperm motility and viability of Tris egg yolk glycerol diluent were better than those of Tris egg yolk DMF (p<0.05). Sperm kinematics showed no difference between homemade and commercial diluents. TKTG demonstrated higher sperm kinematics and motility compared to the other diluents. Recovery rate (RR) values were not significantly different between diluents. Sperm longevity in BoviFree® diluent can best maintain sperm motility for 3 hours of storage (24.65±5.53) and 4 hours of storage (19.69±4.50).
