      Detection of Feline Panleukopenia Virus by Polymerase Chain Reaction (PCR) in Cats with Clinical Symptoms

      Abstract
      Feline Panleukopenia (FP) is a viral disease in cats that is endemic in Indonesia. Diagnosis is generally based on clinical symptoms, but it is often mistaken for infections by other viruses such as feline leukemia virus (FeLV) and feline corona virus (FCoV). Therefore, supportive diagnostic tests are needed, for example rapid antigen tests or detection of viral genetic material using PCR. This study aimed to detect FPV from cats showing clinical symptoms of FP (n=7) and cats with clinical symptoms ambiguous between FP and FIP (n=7). Fourteen archived samples consisting of plasma (n=2), feces (n=3), and rectal swabs (n=9) were analyzed by PCR targeting the viral protein (VP)2 gene. Out of 14 samples tested, 4 showed bands after electrophoresis, but only 1 sample (a rectal swab from a cat with clinical signs of FP) had a band at the expected size of 681 bp. No samples from cats with ambiguous clinical signs between FP and FIP tested positive by PCR, highlighting the importance of PCR as a diagnostic tool to differentiate FP from other diseases with similar symptoms.
       
      Feline Panleukopenia (FP) adalah salah satu penyakit virus pada kucing yang endemik di Indonesia. Diagnosa umumnya berdasarkan gejala klinis, namun sering dikelirukan oleh infeksi virus lainnya seperti feline leukimia virus (FeLV) dan feline corona virus (FCoV). Oleh sebab itu, diperlukan uji diagnostik penunjang seperti uji cepat antigen atau deteksi material genetik virus menggunakan uji PCR. Penelitian ini bertujuan untuk mendeteksi FPV dari kucing yang menunjukkan gejala klinis FP (n=7) dan kucing dengan gejala klinis dubius antara FP dan FIP (n=7). Empat belassampel arsip berupa plasma (n=2), feses (n=3), swab rektal (n=9) dianalisis menggunakan PCR dengan gen target viral protein (VP)2. Dari 14 sampel yang diuji, 4 menunjukkan pita setelah elektroforesis, tetapi hanya 1 sampel (swab rektal dari kucing bergejala FP) yang memiliki pita pada ukuran 681 bp. Tidak ada sampel dari kucing dengan gejala dubius FP dan FIP yang positif PCR, menegaskan pentingnya PCR untuk membedakan FP dari penyakit serupa
