      Potensi Nanopartikel Perak dalam Menekan Infeksi Zucchini yellow mosaic virus Pada Tanaman Mentimun

      Abstract
      Zucchini yellow mosaic virus (ZYMV) merupakan virus utama yang menginfeksi tanaman famili Cucurbitaceae, termasuk mentimun. Umumnya, virus yang ditularkan serangga dikendalikan menggunakan insektisida kimia untuk mengurangi serangga vektor dan infeksi virus. Penelitian ini bertujuan menguji potensi nanopartikel perak (Ag-NP) dalam menekan infeksi ZYMV pada mentimun. Ag-NP disintesis dengan metode biosintesis menggunakan ekstrak daun kelor sebagai reduktor. Ag-NP diaplikasikan dengan cara disemprotkan pada daun sehari sebelum dan sesudah infeksi mekanis ZYMV pada konsentrasi 50 ppm dan 60 ppm. Tanaman kontrol yang sehat dan terinfeksi digunakan sebagai pembanding. Parameter penyakit dan agronomis diamati setiap minggu. Perlakuan Ag-NP menunjukkan mampu memperpanjang masa inkubasi, mengurangi intensitas penyakit serta titer virus dibandingkan dengan kontrol yang terinfeksi ZYMV. Aplikasi Ag-NP setelah infeksi ZYMV menunjukkan penghambatan penyakit yang lebih tinggi dibandingkan dengan perlakuan Ag-NP sebelum infeksi ZYMV dan kontrol. Keefektifan Ag-NP dalam menekan titer ZYMV berkisar antara 57,27% sampai 74,91%, tergantung pada perlakuan. Total klorofil lebih tinggi pada perlakuan Ag-NP dibandingkan kontrol. Aplikasi Ag-NP tidak menghambat perkecambahan benih, indeks vigor, dan pertumbuhan mentimun. Lebih rendahnya titer virus pada perlakuan sebelum dan sesudah inokulasi mengindikasikan Ag-NP bersifat sebagai penginduksi ketahanan tanaman dan antiviral. Ag-NP 50 ppm yang diaplikasikan setelah inokulasi virus merupakan perlakuan terbaik dalam menekan ZYMV.
       
      Zucchini yellow mosaic virus (ZYMV) is a major virus infecting members of the Cucurbitaceae family, including cucumber. Generally, insect-transmitted viruses were controlled using chemical insecticides to reduce vector and virus transmission. This study aimed to evaluate the potential of silver nanoparticles (Ag-NP) in suppressing ZYMV infection on cucumber. The Ag-NP was biosynthesized using moringa leaves extracts as reductor. Ag-NP was applied as foliar spray a day prior and post mechanical infection of ZYMV at concentrations 50 ppm and 60 ppm. Healthy and infected control plants were used as a comparisons. Disease and agronomic parameters were observed every week. Ag-NP treatment was shown to be able to extend the incubation period, reduce disease intensity and virus titer compared to ZYMV-infected controls. Applying Ag-NP after ZYMV infection showed higher disease inhibition than pre-infection application. The effectiveness of Ag-NP in suppressing ZYMV titer ranged from 57.27% to 74.91%, depends on the treatment. Total chlorophyll was higher in Ag-NP treatment compared to control. Ag-NP application did not inhibit seed germination, vigor index, and cucumber growth. The lower virus titer in the pre- and post-inoculation treatments indicated that Ag-NP acted as an inducer of plant resistance and antiviral. Ag-NP 50 ppm applied post-inoculation was the best treatment in suppressing ZYMV.
