      Host Range Of Papaya ringspot virus Isolates Originated from Several Regions in Indonesia

      Abstract
      Papaya ringspot virus (PRSV) is the cause of ringspot disease in papaya plants. The virus has also been reported to infect plants from the Cucurbitaceae family. This study aimed to observe the incidence, severity, and symptom development of ringspot disease caused by PRSV isolates from Medan, Bogor, Denpasar, and Mataram on papaya (var. Callina) and several plants from the Cucurbitaceae family, consisting of cucumber (Cucumis sativus), melon (Cucumis melo), and pumpkin (Cucurbita moschata). A completely randomized design was used in this experiment with two treatment levels, namely plant type and PRSV isolate. Each treatment was repeated three times with 5 experimental units for each replication. Confirmation of PRSV infection was conducted using DAS-ELISA and RT-PCR methods. The types of symptoms that appeared on the leaves after inoculation included mosaic, chlorotic spots, vein clearing, shriveling, curling, lamina thickening, malformation, and shoestring-shaped leaves. The incubation period of the virus in plants ranged from 10 to 28 days after inoculation. Three PRSV isolates, i.e. PRSV-Medan, PRSV-Bogor, and PRSV-Denpasar, were confirmed by DAS-ELISA to infect papaya and Cucurbitaceae plants, while PRSV-Mataram could only infect pumpkin. Sequence analysis showed that PRSV isolates from Bogor, Medan, and Denpasar belonged to PRSV strain P, while isolates from Mataram belonged to strain W.
       
      Papaya ringspot virus (PRSV) adalah penyebab penyakit bercak cincin pada tanaman pepaya. Virus ini juga telah dilaporkan dapat menginfeksi tanaman dari famili Cucurbitaceae. Penelitian ini bertujuan untuk mengamati insidensi, keparahan, dan perkembangan gejala penyakit bercak bercincin yang disebabkan oleh PRSV isolat Medan, Bogor, Denpasar, dan Mataram pada tanaman pepaya (var. Callina) dan beberapa tanaman dari famili Cucurbitaceae, yang terdiri atas mentimun (Cucumis sativus), melon (Cucumis melo), dan labu siam (Cucurbita moschata). Rancangan acak lengkap digunakan dalam percobaan ini dengan dua tingkat perlakuan, yaitu jenis tanaman dan isolat PRSV. Setiap perlakuan diulang sebanyak tiga kali dengan lima unit percobaan untuk setiap ulangan. Konfirmasi infeksi PRSV dilakukan menggunakan metode DAS-ELISA dan RT-PCR. Jenis gejala yang muncul pada daun setelah inokulasi antara lain mosaik, bercak klorotik, vein clearing, mengerut, menggulung, penebalan lamina, malformasi, dan daun berbentuk shoestring. Masa inkubasi virus pada tanaman berkisar antara 10 hingga 28 hari setelah inokulasi. Tiga isolat PRSV, yaitu PRSV-Medan, PRSV-Bogor, dan PRSV-Denpasar, terkonfirmasi dengan DAS-ELISA dapat menginfeksi tanaman pepaya dan tanaman Cucurbitaceae, sedangkan PRSV-Mataram hanya dapat menginfeksi labu kuning. Analisis sekuen menunjukkan bahwa isolat PRSV dari Bogor, Medan, dan Denpasar termasuk ke dalam PRSV strain P, sedangkan isolat dari Mataram termasuk ke dalam strain W.
