      Aktivitas Antibakteri Ekstrak Jintan Hitam (Nigella sativa) Terhadap Bakteri Gram Positif dan Negatif

      Abstract
      Resistansi antimikroba merupakan salah satu permasalahan global. Penggunaan antibiotik yang berlebihan mengakibatkan semakin tingginya kasus resistansi antibiotik. Staphylococcus aureus merupakan bakteri yang mudah mengalami resistansi terhadap antibiotik. Selain bakteri Staphylococcus aureus, bakteri Gram positif lain yaitu Streptococcus serta beberapa strain dari bakteri Gram negatif E. coli dan Salmonella telah resistan terhadap beberapa jenis antibiotik. Oleh karena itu, diperlukan suatu alternatif, salah satunya mencari senyawa-senyawa yang berpotensi sebagai antibakteri dari tumbuhan yang memiliki khasiat obat. Salah satu tumbuhan yang dapat dimanfaatkan sebagai alternatif pengganti antibiotik adalah jintan hitam (Nigella sativa). Penelitian ini bertujuan untuk menguji sifat antibakteri ekstrak jintan hitam terhadap berbagai bakteri secara in vitro serta mengetahui konsentrasi efektif sebagai antibakteri terbaik. Pengujian daya kerja antibakteri ekstrak jintan hitam dilakukan menggunakan metode difusi cakram (disc diffusion method) Kirby Bauer. Hasil penelitian menunjukkan ekstrak jintan hitam dapat menghambat pertumbuhan bakteri Gram positif. Penelitian ini menyimpulkan ekstrak jintan hitam (Nigella sativa) memiliki aktivitas antibakteri yang baik terhadap bakteri Gram positif namun kurang efektif terhadap bakteri Gram negatif. Konsentrasi 100% ekstrak jintan hitam memiliki aktivitas antibakteri paling baik terhadap bakteri Gram positif.
       
      Antimicrobial resistance is a global problem. Excessive use of antibiotics results in increasing cases of antibiotic resistance. Staphylococcus aureus is a bacteria that easily becomes resistant to antibiotics. Apart from Staphylococcus aureus, other Gram-positive bacteria, namely Streptococcus, as well as several strains of Gram-negative bacteria, E. coli and Salmonella, are resistant to several types of antibiotics. Therefore, an alternative is needed, one of which is looking for compounds that have the potential to act as antibacterial from plants that have medicinal properties. One plant that can be used as an alternative to antibiotic is black cumin (Nigella sativa). This study aims to provide the in vitro antibacterial properties of black cumin extract against various bacteria and determine the effective concentration as the best antibacterial. The antibacterial test of black cumin extract was carried out using the Kirby Bauer disc diffusion method. The research results showed that black cumin extract could inhibit the growth of Gram-positive bacteria. This study concluded that black cumin extract had good antibacterial activity against Gram-positive bacteria but is less effective against Gram-negative bacteria. The 100% concentration of black cumin extract has the best antibacterial activity against Gram-positive bacteria.
