Melisopalinologi dan Tumbuhan Pakan Lebah Tanpa Sengat Heterotrigona Itama di Belitung

Tumbuhan, khususnya angiospermae, memiliki hubungan mutualisme dengan berbagai agen polinator, terutama lebah. Di daerah tropis, lebah tanpa sengat merupakan salah satu polinator terpenting yang mengumpulkan nektar dan polen dari berbagai jenis tumbuhan. Data tumbuhan sumber pakan lebah tanpa sengat dapat diperoleh dengan menggunakan analisis melisopalinologi, yaitu metode untuk mengidentifikasi keanekaragaman dan menghitung komposisi polen dalam madu. Peternakan lebah tanpa sengat di Pulau Belitung berlokasi di berbagai tipe penggunaan lahan dan vegetasi. Heterotrigona itama merupakan jenis lebah yang paling banyak diternakkan di Pulau Belitung. Namun, informasi mengenai tumbuhan pakan lebah H. itama di pulau tersebut masih terbatas. Oleh karena itu, penelitian ini bertujuan untuk: (1) mengidentifikasi keanekaragaman tipe polen di dalam madu H. itama yang dikoleksi dari enam lokasi peternakan di Pulau Belitung, (2) menganalisis struktur vegetasi di sekitar peternakan lebah H. itama di enam lokasi peternakan di Pulau Belitung dan (3) menganalisis hubungan antara komposisi tipe polen di dalam madu dan vegetasi di sekitar enam peternakan lebah H. itama di Pulau Belitung.
Penelitian dilaksanakan pada Januari – Desember 2023. Pengoleksian data dilakukan di enam peternakan H. itama yang berlokasi di enam desa di Pulau Belitung yaitu Desa Keciput, Desa Jangkar Asam, Desa Batu Itam, Desa Kelubi, Desa Mekar Jaya, dan Desa Sukamandi. Dari setiap peternakan sampel madu dikoleksi dari tiga koloni. Polen diektraksi dan diasetolisis dari madu, lalu dibuat sediaan preparat untuk diamati ciri morfologinya, diidentifikasi dan dihitung kelimpahannya. Setiap tipe polen kemudian diklasifikasikan berdasarkan kelas frekuensi. Survei vegetasi dilakukan dengan menggunakan metode petak bersarang yang diletakkan mengikuti arah terbang dominan lebah. Jenis tumbuhan yang ditemukan di dalam plot dihitung dan dianalisis menggunakan Indeks Nilai Penting (INP). Keanekaragaman tipe polen di dalam madu dan jenis tumbuhan di sekitar peternakan dianalisis menggunakan Indeks Keanekaragaman Shannon-Wienner (H') dan Indeks Kemerataan Pielou (J'). Analisis kluster berdasarkan koefisien similaritas Jaccard digunakan untuk membandingkan vegetasi dan komposisi polen dalam madu antar peternakan.
Lima puluh tipe polen dari 32 famili tumbuhan ditemukan pada sampel madu H. itama. Myrtaceae, Asteraceae, dan Arecaceae adalah famili yang paling banyak ditemukan dengan masing-masing memiliki enam, lima, dan empat tipe polen. Morfologi polen yang paling sering dijumpai dalam sampel madu adalah aperture 3-colporate (56%), ornamentasi psilate (28%), dan polen berukuran kecil (48%). Sampel madu dari peternakan lebah di enam desa memiliki tipe polen predominan yang berbeda. Tipe polen predominan di dalam madu dari peternakan di Desa Keciput, Desa Batu Itam, dan Desa Mekar Jaya secara berurutan adalah Ilex cymosa, Rhizophora mucronata, dan Mangifera indica. Di Desa Kelubi, terdapat perbedaan tipe polen predominan antar koloni, polen dari Acacia mangium predominan pada satu koloni, sedangkan Syzygium urceolatum predominan pada dua koloni lainnya. Di Desa Jangkar Asam, Melastoma malabathricum merupakan polen predominan di satu koloni, namun merupakan polen sekunder dan minor di dua koloni lainnya. Di Desa Sukamandi, satu koloni memiliki polen predominan dari M. malabathricum, dan koloni lainnya didominasi Cyperus sp.; satu koloni di Desa Sukamandi tidak memiliki polen predominan.
Berdasarkan identifikasi polen di dalam madu diketahui bahwa tumbuhan pakan yang digunakan H. itama di enam peternakan berkisar antara 10 hingga 23 jenis. Hasil identifikasi polen juga menunjukkan bahwa tumbuhan pakan H. itama pada penelitian ini pada umumnya berupa pohon, lebah tersebut juga cenderung mengoleksi bunga dengan mahkota berwarna putih hingga kekuningan, perbungaan majemuk atau tunggal dan memiliki populasi yang besar. Sedangkan berdasarkan survei vegetasi, kekayaan jenis tumbuhan di sekitar peternakan H. itama bervariasi antara 16 hingga 34 jenis, dengan tumbuhan dari habitus pohon yang paling banyak mendominasi di sekitar peternakan.
Hasil survei vegetasi di sekitar peternakan menunjukan bahwa S. urceolatum dan A. mangium memiliki INP tertinggi pada peternakan di Desa Kelubi, hasil ini sejalan dengan tipe polen predominan di lokasi tersebut. Namun, INP tertinggi tidak selalu dapat dikaitkan dengan tipe polen predominan. Di Desa Sukamandi, A. mangium juga memiliki INP tertinggi pada tingkat pancang, namun polen tumbuhan tersebut termasuk dalam important minor pollen. Schima wallichii memiliki INP tertinggi di Desa Keciput dan Desa Jangkar Asam, namun polen dari S. wallichii tergolong polen minor pada madu dari peternakan di Desa Keciput, dan tidak ditemukan pada madu dari peternakan di Desa Jangkar Asam.
Indeks keanekaragaman (H') jenis tumbuhan di sekitar peternakan secara umum sejalan dengan H' tipe polen di dalam madu. Madu dengan H' tipe polen tertinggi dijumpai pada peternakan dengan H' jenis tumbuhan tertinggi. Peternakan lebah di Desa Jangkar Asam memiliki vegetasi dengan nilai H' tertinggi dan menghasilkan madu dengan H' tipe polen tertinggi. Hasil analisis indeks kemerataan (J') menunjukkan pola yang sedikit berbeda. Peternakan lebah di Desa Batu Itam memiliki vegetasi dengan J' tertinggi, namun madu dengan J' tipe polen tertinggi terdapat di Desa Jangkar Asam.
Hasil analisis klaster berdasarkan similaritas menunjukkan bahwa komposisi vegetasi dan tipe polen di dalam madu dari peternakan di Desa Batu Itam dan Desa Mekar Jaya berbeda dengan peternakan lainnya. Peternakan di Desa Batu Itam memiliki vegetasi mangrove dan polen R. mucronata yang tidak ditemukan pada peternakan lainnya. Sedangkan, peternakan di Desa Mekar Jaya memiliki vegetasi dominan berupa taman di pekarangan rumah dan tipe polen predominan M. indica yang juga tidak ditemukan di peternakan lain.
Hasil penelitian ini memberikan data awal mengenai jenis tumbuhan penting untuk H. itama di Pulau Belitung yang dapat digunakan sebagai acuan dalam pengelolaan peternakan lebah H. itama untuk mendukung produksi madu yang berkelanjutan di Pulau Belitung. Plants, especially angiosperms, have a mutualistic relationship with various pollinators, primarily bees. In tropical areas, stingless bees are one of the most important pollinators that collect nectar and pollen from a vast array of plant species. Data on plants foraged by bees could be obtained using melissopalynological analysis, i.e., a method for identifying diversity and calculating pollen composition in honey. Meliponicultures on Belitung Island are located in various types of land use and vegetation. Heterotrigona itama is a stingless bee most commonly used in meliponiculture on Belitung Island. However, information regarding H. itama forage plants on Belitung Island is still limited. Therefore, this study aims to: (1) identify the diversity of pollen types in H. itama honey collected from six meliponicultures in Belitung Island, (2) analyze the vegetation structure around H. itama meliponicultures in six meliponiculture locations in Belitung Island, and (3) analyze the relationship between the composition of pollen types in honey and vegetation around six meliponicultures in Belitung Island.
The study was conducted from January – December 2023. Data was collected at six H. itama meliponicultures located in six villages on Belitung Island, i.e., Keciput, Jangkar Asam, Batu Itam, Kelubi, Mekar Jaya, and Sukamandi. Honey samples were collected from three colonies on each meliponiculture. Pollen was extracted, acetolyzed, and made into microscopic slides to observed morphological characters, identify and calculate pollen abundance. Each type of pollen was classified based on frequency classes. Vegetation analysis was carried out using the nesting plot method, which was placed following the dominant flight direction of the bees. The types of plants found in the plot were calculated and analyzed using the Important Value Index (IVI). The diversity of pollen types in honey and plant types around the meliponiculture were analyzed using the Shannon-Wienner Diversity Index (H') and Pielou Evenness Index (J'). Cluster analysis based on Jaccard similarity coefficient was used to compared vegetation and pollen composition among meliponicultures.
Fifty pollen types from 32 plant families were found in H. itama honey samples. Myrtaceae, Asteraceae, and Arecaceae were the most abundant families, represented by six, five, and four pollen types, respectively. The most common pollen morphologies found in honey samples were 3-colporate aperture (56%), psilate ornamentation (28%), and small pollen size (48%). Honey samples from meliponicultures in six villages had different predominant pollen types. The predominant pollen types in honey from meliponicultures in Keciput, Batu Itam, and Mekar Jaya were Ilex cymosa, Rhizophora mucronata, and Mangifera indica, respectively. In Kelubi honey, there were differences in predominant pollen types between colonies; pollen from Acacia mangium was predominant in one colony, while Syzygium urceolatum was predominant in two other colonies. In Jangkar Asam, Melastoma malabathricum was the predominant pollen in one colony but was secondary and minor in two other colonies. In Sukamandi, one colony had predominant pollen from M. malabathricum, and the other colony was dominated by Cyperus sp.; one colony in Sukamandi had no predominant pollen.
Based on pollen identification in honey, it is known that the forage plants used by H. itama in six meliponicultures range from 10 to 23 species. The results of pollen identification also showed that the forage plants of H. itama in this study were primarily trees. The bees tend to collect flowers with white to yellowish crowns, had compound or single inflorescences that have a large population. Meanwhile, based on vegetation analysis, the richness of plant species around H. itama meliponiculture varied between 16 to 34 species with tree species being the most dominant species surrounding the meliponiculture.
The results of vegetation analysis around the meliponiculture showed that S. urceolatum and A. mangium had the highest IVI in Kelubi; this result is also in line with the predominant pollen type at Kelubi. However, the highest IVI cannot always be associated with the predominant pollen type in the honey. In Sukamandi, A. mangium also had the highest IVI at the sapling level, but the pollen was categorized as important minor pollen. Schima wallichii had the highest IVI in Keciput and Jangkar Asam, but pollen from S. wallichii was a minor pollen in Keciput honey and was not even found in Jangkar Asam honey.
The diversity index (H') of plant species around the meliponiculture was generally in line with the H' type of pollen in honey. Honey with the highest H' type of pollen was found in the meliponiculture with the highest H' type of plant. The meliponiculture in Jangkar Asam had vegetation with the highest H' value, which also produced honey with the highest H' type of pollen. The Evenness index analysis (J') results showed a slightly different pattern. The Batu Itam meliponiculture had the highest J' of vegetation, but honey with the highest J' of pollen type is found in Jangkar Asam Village.
The cluster analysis based on similarity showed that the composition of vegetation and pollen types in honey from Batu Itam and Mekar Jaya differed from other locations. Meliponiculture in Batu Itam had mangrove vegetation and R. mucronata pollen that was not found in other meliponiculture. Meanwhile, meliponiculture in Mekar Jaya had home garden vegetations and predominant pollen from M. indica, which is also not found in other meliponiculture.
This study provide preliminary data on important plant species for H. itama on Belitung Island that can be used as a reference in managing H. itama meliponiculture to support sustainable honey production on Belitung Island.
