Pisang cv Mas Kirana merupakan salah satu kultivar pisang yang banyak
dibudidayakan, terutama di daerah Lumajang karena mempunyai beberapa
keunggulan antara lain warna buah menarik, rasa daging buah manis dan segar,
teksturnya renyah, dan ukuran buah yang kecil. Penyakit kerdil pisang yang
disebabkan oleh infeksi Banana bunchy top virus (BBTV) dianggap sebagai virus
penting yang menyebabkan kehilangan hasil pada tanaman pisang di Asia, Afrika,
dan Pasifik Selatan. Penggunaan bahan tanaman bebas virus direkomendasikan
untuk mencegah penyebaran dan perkembangan penyakit di lapangan. Penelitian
ini melibatkan pengumpulan bahan tanaman dari lapangan untuk digunakan
sebagai eksplan awal untuk kultur jaringan. Penelitian ini bertujuan untuk
mengetahui insidensi penyakit di lapangan dan deteksi BBTV di lapangan
dengan metode PCR. Hasil penelitian menunjukkan bahwa insidensi penyakit
BBTD di lapangan yaitu insidensi penyakit 24,13 34,14% dari total tanaman
yang diamati di setiap blok. Gejala infeksi BBTV yang ditemukan di lapangan
adalah tanaman pisang yang kerdil, daun tumbuh mengumpul atau membentuk
roset di bagian ujung tanaman, dan terjadi klorosis yang disertai adanya nekrosis
di sepanjang tepi daun. DNA berukuran sekitar 240 pb berhasil teramplifikasi
menggunakan pasangan primer spesifik BBTV sebanyak 10 dari total 24 sampel
tanaman. Deteksi dengan menggunakan metode PCR mengkonfirmasi infeksi
BBTV pada tanaman bergejala dan tidak bergejala.
Salah satu kendala dalam budidaya pisang Mas Kirana yaitu jumlah anakan
yang dihasilkan hanya 1-3 anakan. Multiplikasi tunas menjadi aspek penting
untuk keberhasilan kultur jaringan. Semakin banyak tunas yang dihasilkan
diharapkan akan semakin banyak pula persediaan benih untuk produksi. Tujuan
dari penelitian ini yaitu untuk mendapatkan jenis dan konsentrasi sitokinin yang
optimal untuk multiplikasi tunas pisang cv Mas Kirana. Percobaan dilakukan
dengan menggunakan Rancangan Acak Lengkap (RAL) yang terdiri dari 8
perlakuan, yaitu zat pengatur tumbuh sitokinin yang berbeda dengan tingkat
konsentrasi yang berbeda yang ditambahkan ke dalam media multiplikasi (2 mg L- 1 BAP; 4 mg L-1 BAP; 0,25 mg L-1 TDZ; 0,75 mg L-1 TDZ; 4 mg L-1 Kinetin; 5
mg L-1 Kinetin; 2 mg L-1 BAP + 0,25 mg L-1 TDZ dan kontrol). Selain itu, juga
dilakukan konfirmasi apakah planlet hasil kultur jaringan masih terinfeksi BBTV.
Hasil percobaan menunjukkan bahwa jenis dan konsentrasi sitokinin yang berbeda
nyata hanya pada peubah jumlah tunas. BAP 2 mg L-1 mampu menghasilkan
jumlah tunas dengan rata-rata 3,5 tunas. Hasil amplifikasi PCR dari 16 sampel
hasil kultur jaringan yang telah disubkultur dua kali terdapat 7 sampel yang
terkonfimasi BBTV.
Banana bunchy top virus (BBTV) merupakan salah satu virus utama yang
menginfeksi tanaman pisang. Berbagai kultivar pisang yang ditanam di Indonesia
diketahui rentan terhadap BBTV, termasuk cv. Mas Kirana yang merupakan salah
satu pisang komersial yang banyak dibudidayakan. Dilaporkan sebelumnya bahwa
termoterapi merupakan salah satu metode eliminasi virus yang efektif pada
beberapa tanaman. Penelitian ini bertujuan untuk memperoleh suhu termoterapi
yang optimal untuk eliminasi Banana bunchy top virus pada pisang cv. Mas
Kirana. Hasil penelitian menyatakan bahwa suhu 30 oC merupakan suhu dengan
persentase hidup tertinggi (100%), persentase membentuk tunas sebesar 70%, dan
rata-rata jumlah nodul yang dihasilkan adalah 2,33. Hasil konfirmasi BBTV
dengan PCR menyatakan bahwa suhu 25 oC, 30 oC, 35 oC, dan 40 oC belum
mampu mengeliminasi BBTV melalui metode termoterapi kultur jaringan. Hal ini
diduga karena ukuran nodul yang digunakan terlau besar dan subkultur masih dua
kali. Banana cv Mas Kirana is a widely cultivated banana variety, especially in
the Lumajang, because it has several advantages, including attractive fruit color,
sweet and fresh pulp taste, crunchy texture, and small fruit size. Banana bunchy
top disease caused by Banana bunchy top virus (BBTV) infection is considered
the most important virus affecting yield losses of banana plants in Asia, Africa,
and the South Pacific. Using virus-free plant material is recommended to manage
disease spread and development in the field. The research involved collecting
plant material from the field to be used as an initial explant for tissue culture. This
research aims to determine the incidence of disease in the field and the detection
of BBTV in the field by PCR method. The results showed that the incidence of
BBTV in the Sukamantri Research Station ranged from 24,13- 34,14% of the total
plants observed in each block. Symptoms of BBTV infection found in the field are
dwarfed banana plants, leaves that grow clumped or form a rosette at the tip of the
plant, and chlorosis with necrosis along the edges of the leaves. DNA bands
measuring about 240 bp were successfully amplified using BBTV-specific primer
pairs in 10 of 24 plant samples. Detection using the PCR method confirmed the
infection of BBTV on symptomatic and asymptomatic plants.
One of the problems in the cultivation of banana cv Mas Kirana is that the
number of shoots produced is only 1-3. Shoot multiplication is an essential aspect
of the success of tissue culture. The more shoots produced, the more the supply of
seeds is expected to increase. This study aimed to determine the optimal type and
concentration of cytokinin for Mas Kirana micropropagation. The experiment was
conducted using a completely randomized design consisting of 8 treatments, i.e.
different cytokinin growth regulator in different concentration levels that added to
multiplication media (2 mg L-1 BAP; 4 mg L-1 BAP; 0.25mg L-1 TDZ; 0.75 mg L- 1 TDZ; 4 mg L-1 Kinetin; 5 mg L-1 Kinetin; 2 mg L-1 BAP + 0.25 mg L-1 TDZ and
control). Furthermore, it was also confirmed whether the tissue culture planlets
were still infected with BBTV. The results showed that the type and concentration
of cytokinin were significantly different only on the variable number of shoots.
BAP 2 mg L-1 produces the number of shoots with an average of 3,5 shoots. PCR
amplification results from 16 tissue culture samples that were subcultured twice;
seven confirmed BBTV.
Banana bunchy top virus (BBTV) is one of the major viruses that infect
banana plants. Various banana cultivars grown in Indonesia are known to be
susceptible to BBTV, including cv. Mas Kirana is one of the most one of the most
widely cultivated commercial bananas. It was previously reported that
thermotherapy is one of the effective virus elimination methods in several crops.
This study aims to obtain the optimal thermotherapy temperature for eliminating
the Banana bunchy top virus in banana cv. Mas Kirana. The results stated that 30
C was the temperature with the highest percentage of survival (100%), the
percentage of nodule formation was 70%, and the average number of nodules
produced was 2.33. The results of BBTV confirmation by PCR showed that
temperatures of 25 oC, 30 oC, 35 oC, and 40 oC could not eliminate BBTV through
the tissue culture thermotherapy method. This is probably because the size of the
nodules used was too large, and the subculture was still two times.
