      Histopathology of the Spleen from Streptozotocin-Induced Diabetic Rats Treated with Freeze-Dried Cocos nucifera

      Abstract
      Diabetes mellitus (DM) is widely recognized as one of the leading causes of death and disability worldwide. This study aims to study the histopathological features of the spleen of diabetic rats treated with freeze-dried young coconut compared with glibenclamide as a positive standard for DM treatment. Nine male Sprague-Dawley rats were divided into five treatment groups which were all induced diabetes using streptozotocin at a dose of 55 mg/kg, except for the normal group (KN) which was only given 1 mL of distilled water. The diabetes group (KD) was given 1 mL of distilled water, the positive control (DG) was given glibenclamide 0.6 mg/kg BW, DSK3.5 was given 3,5 g/kg BW freeze-dried young coconut, and the DSK7 group was given 7 g/kg BW freeze-dried young coconut orally as a feed supplement. All rats were euthanized, and spleens were sampled to make histopathological preparations using hematoxylin-eosin staining. DSK3.5 showed improvement, namely a decrease in total lymphoid follicles and an increase in size and density. Although not significantly different, these findings indicate that freeze-dried young coconut has an effect similar to glibenclamide, allowing it to be considered as an option for DM therapy.
       
      Diabetes mellitus (DM) diakui secara luas sebagai salah satu penyebab utama kematian dan kecacatan di seluruh dunia. Penelitian ini bertujuan untuk mempelajari gambaran histopatologi limpa tikus diabetes yang diberi perlakuan kelapa muda kering-beku dengan konsentrasi berbeda, dan dibandingkan dengan glibenklamid sebagai standar positif pengobatan DM. Sembilan ekor tikus jantan Sprague-Dawley dibagi menjadi lima kelompok perlakuan yang seluruhnya diinduksi diabetes menggunakan streptozotocin (STZ) dosis 55 mg/kg, kecuali kelompok normal (KN) yang hanya diberi akuades 1 mL. Keempat kelompok lainnya yaitu kelompok diabetes (KD) yang diberi akuades 1 mL, kontrol positif (DG) diberi glibenclamide 0,6 mg/kg BB, DSK3.5 diberi air kelapa kering-beku 3,5 g/kg BB, dan kelompok DSK7 diberi air kelapa kering-beku 7 g/kg BB per oral sebagai supplemen. Seluruh perlakuan dilaksanakan sehari sekali selama 45 hari. Selanjutnya seluruh tikus dieutanasi, limpa disampling untuk dibuat sediaan histopatologi dan diwarnai dengan hematoxilin-eosin. Tikus DM yang diberi air kelapa muda kering-beku 3,5 g/kg BB (DSK3.5) menunjukkan limpa yang membaik yaitu folikel limfoid total berkurang namun ukuran dan kepadatan folikel limfoid meningkat. Meskipun tidak berbeda signifikan, temuan ini menunjukkan bahwa air kelapa muda kering-beku memiliki pengaruh yang mirip dengan glibenklamid sehingga dapat dipertimbangkan sebagai pilihan untuk terapi DM.
