dc.description.abstract | Kalium merupakan makronutrien penting untuk pertumbuhan dan
perkembangan tanaman. Secara umum, sebagian besar (90-98%) kalium di alam
dalam bentuk tidak tersedia atau tidak dapat diserap oleh tanaman sehingga
diperlukan metode untuk mempercepat pelarutan K, salah satunya menggunakan
bakteri pelarut kalium. Penelitian ini bertujuan mengetahui populasi, melakukan
isolasi, dan identifikasi bakteri pelarut kalium dari berbagai lahan di Dadahup,
Kabupaten Kapuas, Kalimantan Tengah. Hasil penelitian menunjukkan bahwa
populasi bakteri pelarut kalium tertinggi pada lokasi lahan sawah bera 1 tahun atau
tanam tetapi kena banjir dengan populasi 7,38x105 CFU/g tanah. Hasil uji
kualitatif memperoleh dua isolat bakteri pelarut kalium tertinggi yaitu PK4L1 dan
PK2L2 dengan indeks pelarutan K sebesar 1,50 dan 1,67, sedangkan uji kuantitatif
dari isolat terpilih yaitu PK4L1 sebesar 3,66 cmol(+)/kg dan PK2L2 sebesar 3,73
cmol(+)/kg dengan masa inkubasi 20 hari. Berdasarkan uji patogenitas berupa uji
hipersensitif menunjukkan isolat PK4L1 dan PK2L2 bersifat tidak patogen
terhadap tanaman, sedangkan uji hemolisis menunjukkan isolat PK4L1 bersifat
patogen terhadap binatang dan PK2L2 bersifat tidak patogen terhadap binatang.
Identifikasi isolat secara molekuler menggunakan gen 16S rRNA berdasarkan
hasil BLAST pada NCBI dan analisis pohon filogenetik menyatakan bahwa isolat
PK4L1 memiliki kemiripan 98,81% dengan spesies Bacillus pumilus strain ATCC
7061 ICBB 9960. Sementara isolat PK2L2 memiliki kemiripan dengan spesies
Bacillus pseudomycoides strain NBRC 101232 ICBB 9961 dengan tingkat
kemiripan 98,34%. | id |
dc.description.abstract | Potassium is an important macronutrient for plant growth and development.
In general, most (90-98%) of potassium in nature in its form is unavailable or cannot
be absorbed by plants so a method is needed to accelerate the dissolution of K, one
of which uses potassium solvent bacteria. This study aims to determine the
population, isolate, and identify potassium solvent bacteria from various fields in
Dadahup, Kapuas District, Central Kalimantan. The results showed that the
population of potassium solvent bacteria is the highest at the location of fallow
rice fields for 1 year or planted but was flooded with a population of 7,38x105
CFU/g of soil. The qualitative test results obtained two isolates of the highest
potassium solvent bacteria, namely PK4L1 and PK2L2 with a K dissolving index
of 1,50 and 1,67, while the quantitative test of the selected isolates was PK4L1 of
3,66 cmol(+)/kg and PK2L2 of 3,73 cmol(+)/kg with an incubation period of 20
days. Based on pathogenicity tests in the form of hypersensitive tests, it shows
that PK4L1 and PK2L2 isolates are not pathogenic to plants, while hemolysis tests
show that PK4L1isolates are pathogenic to animals and PK2L2 is not pathogenic
to animals. Molecular identification of isolates using the 16S rRNA gene based on
BLAST results on NCBI and phylogenetic tree analysis stated that the PK4L1
isolate had asimilarity 98,81% resemblance to the species Bacillus pumilus strain
ATCC 7061 ICBB 9960. Meanwhile, the PK2L2 isolate has similarities with the
species Bacillus pseudomycoides strain NBRC 101232 ICBB 9961 with a
similarity rate of 98,34%. | id |
