Toksisitas Ekstrak Etanol Kulit Kayu Sonokeling (Dalbergia latifolia Roxb.) terhadap Larva Udang Artemia salina Leach.
Abstract
Pohon sonokeling merupakan pohon asli Indonesia yang biasa digunakan untuk furnitur dan bangunan. Sonokeling juga dimanfaatkan sebagai obat tradisional oleh masyarakat. Kulit sonokeling berpotensi sebagai antikanker. Penelitian ini bertujuan untuk menentukan rendemen dan toksisitas ekstrak kulit kayu sonokeling hasil ekstraksi dengan berbagai variasi konsentrasi etanol terhadap larva udang A. salina Leach, serta menganalisis karakteristik fitokimia secara kualitatif maupun dari profil spektrum Fourier Transform Infrared Spectroscopy (FTIR) ekstrak tersebut. Metode ekstraksi yang dilakukan adalah metode sokletasi untuk pelarut etanol 50% (E50), 75% (E75), 100% (E100) dan metode maserasi untuk pelarut air (E0) dan etanol 25% (E25). Hasil penelitian ini menunjukkan bahwa semakin rendah konsentrasi etanol menghasilkan rendemen yang semakin tinggi. Rendemen ekstrak terlarut air (E0) tertinggi (18,57%), diikuti oleh ekstrak terlarut E25 (12,59%), E75 (4,97%), E50 (4,47%), dan E100 (3,44%). Hasil pengujian toksisitas menunjukkan bahwa ekstrak terlarut E100 merupakan ekstrak yang paling toksik dan tergolong toksisitas sedang (IC50 153,815 ppm), sedangkan ekstrak terlarut E0 dengan rendemen tertinggi memiliki toksisitas yang tergolong rendah (IC50 792,501 ppm). Hasil analisis fitokimia dan FTIR mengkonfirmasi keberadaan senyawa flavonoid, fenolhidrokuinon, tanin, saponin, steroid, dan triterpenoid di dalam ekstrak kulit kayu sonokeling. Sonokeling tree is a native Indonesian tree which is commonly used for furniture and buildings. Sonokeling is also used as traditional medicine by the community. Sonokeling has potential as an anticancer. This study aimed to determine the yield and toxicity of sonokeling bark extract from the extraction using the various concentrations of ethanol against shrimp larvae of Artemia salina Leach and analyze the phytochemical characteristics of the extract. The extraction used the soxhletation method for solvents of ethanol 50% (E50), 75% (E75), 100% (E100) and maceration method for solvents of water (E0) and 25% ethanol (E25). The results of this study indicated that the lower the ethanol concentration, the higher the yield. The highest yield was the water soluble extract (E0) (18.57%), followed by the ethanol soluble extracts of E25 (12.59%), E75 (4.97%), E50 (4.47%), and E100 (3.44%). The E100 soluble extract was the most toxic extract and classified as moderate toxicity (IC50 153.815 ppm), while the E0 soluble extract had low toxicity (IC50 792.501 ppm). The results of phytochemical analysis and FTIR spectrum confirmed the presence of flavonoid compounds, phenolhydroquinones, tannins, saponins, steroids, and triterpenoids in the sonokeling bark extracts.
