      Profil Eritrosit Sapi Peranakan Ongole yang Disimpan Selama 48 Jam dalam Antikoagulan Heparin

      Abstract
      Erythrocyte is one of the main parameters in blood tests. The blood sample that tested is sometimes damaged, so it is necessary to add an anticoagulant to keep the blood intact. This study was conducted to determine the profile of erythrocytes in blood samples of Ongole Crossbred Cattle with heparin added and different storage times. This study used 9 Ongole Crossbred Cattle aged 4-5 years. Blood samples were collected through the coccygeal vein and stored in a vacutainer tube containing the anticoagulant heparin. Samples were stored in the refrigerator and analyzed for the total parameters of RBC, HGB, HCT, MCV, MCH, and MCHC at 0-3, 3, 6, 9, 12, 24, and 48 hours. The results showed that the total number of RBC, HGB, HCT, MCV, MCH and MCHC fluctuated during the observation time. The results of the examination of the total number of RBC, HGB, HCT, MCV, MCH and MCHC values of PO cattle added with heparin anticoagulant did not show significantly different results (P> 0.05) in each hour of observation. This shows that the addition of the anticoagulant heparin to Ongole Croosbred Cattle blood samples can be used for up to 48 hours.
       
      Eritrosit merupakan salah satu parameter utama dalam pemeriksaan darah. Sampel darah yang diuji terkadang mengalami kerusakan sehingga perlu ditambahkan antikoagulan agar kondisi darah tetap utuh. Penelitian ini dilakukan untuk mengetahui profil eritrosit Sapi Peranakan Ongole (PO) yang ditambahkan heparin dengan waktu penyimpanan berbeda. Penelitian ini menggunakan 9 ekor sapi PO yang berumur 4-5 tahun. Sampel darah dikoleksi melalui vena coccygea dan disimpan dalam tabung vaccutainer berisi antikoagulan heparin. Sampel disimpan dalam lemari pendingin dan dilakukan analisa terhadap parameter total RBC, HGB, HCT, MCV, MCH, dan MCHC pada waktu 0-3, 3, 6, 9, 12, 24, dan 48 jam. Hasil penelitian menunjukkan bahwa jumlah total RBC, HGB, HCT, MCV, MCH dan MCHC mengalami fluktuasi selama waktu pengamatan. Hasil pemeriksaan jumlah total nilai RBC, HGB, HCT, MCV, MCH dan MCHC sapi PO yang ditambahkan antikoagulan heparin tidak menunjukkan perbedaan yang nyata (P>0,05) dalam setiap jam pengamatan. Hal ini menunjukkan bahwa penambahan antikoagualan heparin pada sampel darah sapi PO dapat digunakan hingga rentang waktu sampai 48 jam.
