Kapasitas Androgenesis Kultur Antera pada Genotipe Terung (Solanum melongena L.) dengan Kandungan Asam Amino dan Fenol Endogen Berbeda

Androgenesis memiliki peran penting dalam mempersingkat tahapan untuk
menghasilkan galur murni. Pemberian cekaman diperlukan untuk menginduksi
androgenesis. Namun, pemberian cekaman berpotensi menyebabkan kerusakan
oksidatif, sehingga dibutuhkan senyawa yang berperan sebagai antioksidan untuk
mengurangi kerusakan oksidatif dan meningkatkan keberhasilan androgenesis.
Perbedaan warna buah pada terung dapat menandakan perbedaan komposisi
senyawa kimianya, termasuk asam amino dan fenol pada anteranya yang memiliki
aktivitas antioksidan. Prolina merupakan salah satu asam amino yang diketahui
dapat meningkatkan androgenesis. Floroglusinol merupakan senyawa fenol yang
sering digunakan pada kultur in vitro. Namun, studi tentang potensi prolina dan
floroglusinol dalam menginduksi androgenesis terung dengan kandungan fenol
dan asam amino yang berbeda belum pernah diteliti. Oleh karena itu, penelitian
ini bertujuan menguji pengaruh prolina dan floroglusinol dalam menginduksi
androgenesis genotipe terung yang memiliki warna buah yang berbeda dengan
kandungan asam amino dan fenol berbeda pada anteranya.
Bahan tanaman yang digunakan yaitu antera dari empat genotipe terung
(Solanum melongena L.) dengan warna buah, serta kandungan asam amino dan
fenol yang berbeda pada anteranya, yaitu 'Ratih Hijau', 'Ratih Ungu', 'Ratih Putih',
dan 'Antaboga' dengan warna buah ungu. Sebelum antera diisolasi, kuncup bunga
diberi perlakuan cekaman suhu rendah 4 ºC selama 24 jam. Prolina diberikan
selama satu minggu pertama kultur dengan konsentrasi 0, 1, 2, dan 4 mM dan
floroglusinol diberikan setelah satu minggu kultur dengan konsentrasi 0, 100, 200,
dan 400 µM. Proses pembelahan sporofitik diamati setiap minggu dengan
membuat sediaan mikrospora yang diberi pewarna DAPI menggunakan
mikroskop flouresens. Embrio yang terbentuk dihitung, disubkultur hingga
membentuk planlet yang siap untuk aklimatisasi, dan dilakukan analisis ploidi.
Antera yang digunakan sebagai sumber eksplan yang mengandung populasi
mikrospora stadia uninukleat akhir di atas 50% adalah antera dari kuncup bunga
dengan panjang kelopak berkisar antara 2,20-2,40 cm, panjang mahkota 1,00-1,20
cm, dan panjang mahkota tidak melebihi panjang tabung kelopak. Warna antera
juga dapat digunakan untuk menunjukkan antera yang responsif, yaitu warna hijau
untuk „Ratih Hijau‟, kuning pucat untuk „Ratih Putih‟, dan hijau kekuningan
untuk „Ratih Ungu‟ dan „Antaboga‟. Mikrospora yang berhasil diinduksi ke jalur
pembelahan sporofitik dapat ditandai dengan terbentuknya dua inti simetris pada
minggu ke-2 kultur, 5-6 inti pada minggu ke-3, dan banyak inti (pre-globular)
pada minggu ke-4.
Penambahan prolina dapat meningkatkan jumlah embrio pada 'Ratih Hijau'
yang memiliki total asam amino endogen rendah pada antera. Jumlah embrio
tertinggi ditemukan pada „Ratih Hijau‟ dengan penambahan prolina 4 mM.
Penambahan prolina dan kombinasinya dengan floroglusinol cenderung dapat
meningkatkan jumlah embrio pada „Ratih Ungu‟ yang memiliki total fenol dan
asam amino endogen yang sedang pada anteranya. Jumlah embrio tertinggi pada
'Ratih Ungu' ditemukan pada kultur dengan penambahan 4 mM prolina saja
maupun kombinasinya dengan 100 µM floroglusinol. Genotipe yang
menghasilkan embrio paling sedikit adalah 'Ratih Putih', yaitu hanya satu embrio
abnormal yang terbentuk pada kultur dengan penambahan 4 mM prolina yang
dikombinasikan dengan 100 µM floroglusinol. Penambahan kedua senyawa
tersebut tidak mempengaruhi pembentukan embrio pada „Antaboga‟ yang
memiliki total fenol dan asam amino yang lebih tinggi dibandingkan dengan
genotipe lainnya. Embrio pada „Antaboga‟ terbentuk secara acak pada beberapa
konsentrasi prolina dan floroglusinol serta tidak menunjukkan pola khusus.
Embrio yang dihasilkan dapat berupa embrio normal dan embrio abnormal.
Hasil pengamatan histologi embrio menunjukkan bahwa embrio normal memiliki
bagian shoot apical meristem dan root apical meristem yang tumbuh ke dua arah
yang berlawanan, sedangkan embrio abnormal memiliki root apical meristem
yang normal, namun memiliki shoot apical meristem yang tidak normal atau
bahkan tidak terbentuk. Analisis ploidi yang dilakukan pada tanaman yang berasal
dari embrio normal hasil kultur antera menunjukkan bahwa tanaman yang
dihasilkan merupakan tanaman haploid. Androgenesis plays an important role in shortening the steps to create the
pure line. Stress is required to induce androgenesis. However, stress may cause
oxidative damage, so compounds that act as antioxidants are needed to reduce
oxidative damage and increase androgenesis. Differences in fruit color in eggplant
can indicate differences in the composition of chemical compounds, including
amino acids and phenols in the anthers, which have antioxidant activity. Proline is
one of the amino acids known to increase androgenesis. Phloroglucinol is a
phenolic compound that is often used in in vitro culture. However, studies on the
potential of proline and phloroglucinol in inducing androgenesis of eggplant with
different phenol and amino acid content have not been studied. Therefore, this
study aimed to examine the effect of proline and phloroglucinol in inducing
androgenesis of eggplant genotypes that had different fruit colors, with different
amino acid and phenol content in the anthers.
The plant materials used were anthers of four eggplants (Solanum
melongena L.) genotypes with different fruit colors, amino acid, and phenol
content, namely 'Ratih Hijau' with green fruit, 'Ratih Ungu' with purple fruit,
'Ratih Putih' with white fruit, and 'Antaboga' with purple fruit. Before the anthers
were isolated, the flower buds were treated with low-temperature stress of 4 ºC for
24 hours. Proline was given during the first week of culture at 0, 1, 2, and 4 mM.
Phloroglucinol was given after one week of culture at 0, 100, 200, and 400 µM.
The process of the sporophytic division was observed every week by making the
preparations of microspore stained with DAPI and observed using a fluorescent
microscope. The formed embryos were counted, subcultured to form plantlets
ready for acclimatization, and ploidy analysis was performed.
The anthers used as the source of explants containing a population of
microspores in the late uninucleate stage above 50% were the anthers of flower
buds with sepal lengths 2,20-2,40 cm, petal length 1,00-1,20 cm, and petal length
not exceeding the length of the calyx tube. Anther colors can also be used to
indicate the responsive anther, namely green for 'Ratih Hijau', pale yellow for
'Ratih Putih', and yellowish-green for 'Ratih Purple' and 'Antaboga'. Microspores
that were successfully induced to the sporophytic pathway can be characterized by
the formation of two symmetrical nuclei at the 2nd week of culture, 5-6 nuclei at
the 3rd week, and many nuclei (pre-globular) at the 4th week.
The addition of proline can increase the number of embryos in 'Ratih Hijau'
with a low total endogenous amino acid in the anther. The highest number of
embryos was found in 'Ratih Hijau' with the addition of 4 mM proline. The
addition of proline and its combination with phloroglucinol tends to increase the
number of embryos in 'Ratih Ungu' with moderate total phenols and endogenous
amino acids in the anther. The highest number of embryos in 'Ratih Ungu' was
found in cultures with the addition of 4 mM proline only, and its combination
with 100 µM phloroglucinol. The genotype that produced the fewest embryos was
'Ratih Putih'. Only one abnormal embryo was formed in culture with the addition
of 4 mM proline combined with 100 µM phloroglucinol. The addition of these
two compounds did not affect the formation of embryos in Antaboga, which had
higher total phenols and amino acids compared to other genotypes. Embryos in
'Antaboga' formed randomly at several concentrations of proline and
phloroglucinol and did not show a specific pattern.
The embryos can be either normal embryos or abnormal embryos. The
results of histological observations of embryos showed that normal embryos had
shoot apical meristem and root apical meristem that grew in two opposite
directions. In contrast, abnormal embryos have normal root apical meristem but
have an abnormal shoot apical meristem or were not even formed. Ploidy analysis
on plants derived from a normal embryo from anther culture showed that the plant
was haploid.
